摘要

目的 建立同时测定橘红颗粒中芸香柚皮苷、柚皮苷和橙皮苷含量的高效液相色谱法.方法 色谱柱为Agilent 5TC-C18(2)柱(250 mm×4.6 mm,5 μm),流动相为乙腈-0.1%醋酸水溶液(梯度洗脱),流速为 1.0 mL/min,检测波长为 280 nm,柱温为 30℃,进样量为 10 μL.结果 芸香柚皮苷、柚皮苷和橙皮苷进样量分别在 0.034 8~0.348 μg(r = 0.999 5)、1.047~10.47 μg(r = 0.999 4)、0.185 9~1.859 μg(r = 0.999 7)范围内与峰面积线性关系良好;精密度、稳定性、重复性试验结果的RSD均不大于 2.0%;平均加样回收率分别为 98.24%,96.29%,97.83%,RSD分别为 1.16%,1.08%,2.34%(n = 6).结论 所建立的方法操作简便、重复性好、结果准确,可用于同时测定橘红颗粒中上述 3 种黄酮类成分的含量.

Abstract

Objective To establish a high-performance liquid chromatography(HPLC)method for simultaneous determination of narirutin,naringin and hesperidin in Juhong Granules.Methods The chromatographic column was Agilent 5TC-C18(2)column(250 mm×4.6 mm,5 μm),the mobile phase was acetonitrile-0.1%acetic acid aqueous solution(gradient elution),the flow rate was 1.0 mL/min,the detection wavelength was 280 nm,the column temperature was 30℃,and the injection volume was 10 μL.Results The linear ranges of narirutin,naringin and hesperidin were 0.034 8-0.348 μg(r = 0.999 5),1.047-10.47 μg(r = 0.999 4),0.185 9-1.859 μg(r = 0.999 7),respectively.The RSDs of precision,stability and repeatability tests were all lower than 2.0%.The average recovery rates of the above three flavonoids were 98.24%,96.29%,97.83%,with the RSDsof 1.16%,1.08%,2.34%(n = 6)respectively.Conclusion The established method is easy,repeatable and accurate,which can be used to simultaneously determine the contents of the above three flavonoids in Juhong Granules.
