Content Determination of Five Flavonoids in Banzhilian Formula Granules by QAMS
Objective To establish a quantitative analysis of multi-components by single-marker(QAMS)method for the simultaneous determination of five flavonoids(scutellarin,baicalin,scutellarein,naringin,and apigenin)in Banzhilian Formula Granules.Methods The Banzhilian Formula Granules were extracted by ultrasound with 80%methanol,the chromatographic column was Agilent Eclipse XDB C18 column(250 mm×4.6 mm,5 μm),the mobile phase was formic acid solution(pH 2.5)-methanol(65∶35,V/V),the flow rate was 0.8 mL/min,the column temperature was 30℃,the detection wavelength was 280 nm,and the injection volume was 10 μL.With scutellarin as the reference,the relative retention time and relative correction factors of baicalin,scutellarein,naringin and apigenin were respectively established,and the contents of five flavonoids in nine batches of Banzhilian Formula Granules were determined.Results The linear ranges of baicalin,scutellarein,naringin,apigenin,and scutellarin were 1.58-50.63 μg/mL(r = 0.999),6.33-101.25 μg/mL(r = 0.999),1.57-50.25 μg/mL(r = 0.998),1.45-46.50 μg/mL(r = 0.998),and 28.14-900.50 μg/mL(r = 0.997),respectively.The average recoveries of baicalin,scutellarein,naringin,apigenin,and scutellarin were 97.44%,96.78%,100.59%,96.01%,and 95.33%with RSDs of 1.79%,1.67%,2.86%,1.79%,and 1.35%(n = 6),respectively.The RSDs of precision,repeatability,and stability test results were all lower than 3.00%(n = 6).The RSDs of relative retention time and relative correction factor of baicalin,scutellarein,naringin,and apigenin measured by different high-performance liquid chromatography systems and columns were all lower than 3.00%(n = 9).The relative deviation of the content of five flavonoids in nine batches of Banzhilian Formula Granules determined by the external standard method and the QAMS method was not higher than 1.48%(n = 9).Conclusion This method is simple,reliable,and accurate,which can be used for the simultaneous determination of five flavonoids in Banzhilian Formula Granules.
