Effect of Guben Anshen Formula on Sex Hormone Levels and Ovarian Estrogen Receptor Protein Expression Levels in Perimenopausal Syndrome Model Rats
Objective To investigate the effect of Guben Anshen Formula on sex hormone levels and ovarian estrogen receptor protein expression levels in perimenopausal syndrome(PMS)model rats.Methods A total of 60 female SD rats with regular estrous cycles were selected and intraperitoneally injected with 160 mg/kg deoxyethylcyclohexene(VCD)to replicate the PMS model in rats.After successful modeling,60 female rats were randomly divided into the model group(group B,equal volume of normal saline),traditional Chinese medicine group[group C,0.56 g/(kg·d)of Gengnian'an Tablets],Western medicine group[group D,0.1 mg/(kg·d)of Estradiol Valerate Tablets],and Guben Anshen Formula high-,medium-,and low-dose groups(groups E1,E2,E3,18.0,9.0,4.5 g/(kg·d)],with 10 rats in each group.Another 10 female rats were selected as the normal group(group A,equal volume of normal saline).After four weeks of gavage administration,the uterus and bilateral ovaries of rats in each group were removed and weighed with wet weight to calculated the uterine and ovarian indexes.Enzyme-linked immunosorbent assay(ELISA)was used to determine the levels of serum follicle stimulating hormone(FSH),luteinizing hormone(LH),and estradiol(E2).Western blot was used to detect the expression levels of estrogen receptor α(ERα),ERβ and G-protein coupled estrogen receptor 1(GPER1)proteins in ovarian tissues.Results Compared with those in group B,the uterine and ovarian indexes in groups C,D,and E1 were significantly higher(P<0.01);the levels of serum FSH and LH were significantly lower(P<0.01),while the level of E2 was significantly higher in groups C,D,and E1(P<0.01);the expression levels of ERα,ERβ,and GPER1 proteins in the ovarian tissues in groups C,D,and E1 were significantly higher(P<0.01),the expression levels of ERα protein in the ovarian tissues in groups E2 and E3 were significantly higher(P<0.05),and the expression levels of GPER1 protein in the ovarian tissues in group E2 were significantly higher(P<0.05).Conclusion The mechanism of Guben Anshen Formula in the treatment of PMS model rats may be related to its influence on sex hormone secretion and the increase of expression levels of ovarian estrogen receptor proteins.
