Determination of Concentration of Voriconazole in Human Plasma by HPLC
Objective To establish a high-performance liquid chromatography(HPLC)method for the determination of the concentration of voriconazole in human plasma.Methods Acetonitrile protein precipitation method was used to process blood samples.The chromatographic column was InsertSustain C18 column(250 mm×4.6 mm,5 μm),the mobile phase was methanol-acetonitrile-phosphate buffer(33∶28∶39,V/V/V),the flow rate was 1.0 mL/min,the column temperature was 30℃,the detection wavelength was 262 nm,and the injection volume was 20 μL.Results The linear range of voriconazole was 0.1-8 μg/mL(R2=0.999 8,n=8).The limit of quantification was 0.07 μg/mL.The RSDs of the intra-day and inter-day precision,freeze-thaw,and long-term stability test results for samples with low-,medium-,and high-concentrations of 0.125,1,8 μg/mL were all lower than 5% .The extraction recovery rates were(97.32±1.71)%,(97.61±1.22)%,and(96.38±0.84)%,respectively.In the plasma of 60 patients diagnosed with invasive fungal infections,the concentration of voriconazole was in the range of 0.278-9.129 μg/mL.Conclusion The established method is simple,rapid,and has good specificity,which is suitable for monitoring the blood concentration of voriconazole.
