炒榧子仁高效液相色谱特征图谱研究

目的 建立炒榧子仁的高效液相色谱(HPLC)特征图谱.方法 色谱柱为Thermo Hypersil Gold C18 柱(250 mm×4.6 mm,5µm),流动相为乙腈-0.1%磷酸水溶液(梯度洗脱),流速为 1.0 mL/min,检测波长为 203 nm,柱温为 30℃,进样量为 10µL.建立 13 批药材饮片样品的HPLC特征图谱,采用中药色谱指纹图谱相似度评价软件(2012 版)进行相似度评价,确定特征峰,并指认共有峰.结果 确定了 7 个特征峰,其相对保留时间的RSD均小于 1%,相对峰面积的RSD为 7.31%~70.45%;指认了其中 2 个,分别为亚油酸和油酸;13 批药材饮片样品的相似度为 0.827~0.990(仅 2 批低于 0.900).结论 该方法操作简便、可行,结果稳定、准确,有助于完善各地炒榧子仁饮片的炮制规范,提高其质量标准.
HPLC Characteristic Chromatograms of Torreyae Semen Tostum
Objective To establish a high-performance liquid chromatography(HPLC)characteristic chromatograms of Torreyae Semen Tostum.Methods The chromatographic column was the Thermo Hypersil Gold C18 column(250 mm×4.6 mm,5 µm),the mobile phase was acetonitrile-0.1% phosphoric acid aqueous solution(gradient elution),the flow rate was 1.0 mL/min,the detection wavelength was 203 nm,the column temperature was 30℃,and the injection volume was in the range of 10 µL.The HPLC characteristic chromatograms of 13 batches of medicinal material decoction pieces were established,the Similarity Evaluation Software for Chromatographic Fingerprint of Traditional Chinese Medicine(2012 Version)was used for similarity evaluation,the characteristic peaks were marked and the common peaks were identified.Results Seven characteristic peaks were marked,the RSDs of relative retention time were lower than 1%,and the RSDs of relative peak area were in the range of 7.31%-70.45% .Two of them were identified,namely linoleic acid and oleic acid.The similarity of 13 batches of medicinal material decoction pieces was in the range of 0.827-0.990,with only two batches below 0.900.Conclusion This method is simple,feasible,stable and accurate,which can help to improve the processing specifications and quality standards of Torreyae Semen Tostum Decoction Pieces in various producing areas.
