Effect of Gecko Peptides Mixture on the Proliferation and Apoptosis of Human Non-Small Cell Lung Cancer H1299 Cells
Objective To investigate the effect of gecko peptides mixture(GPM)on the proliferation and apoptosis of human non-small cell lung cancer(NSCLC)H1299 cells.Methods The experiment was conducted with the normal control group(equal volume of basic culture medium),high-dose and low-dose GPM groups(20,10 mg/mL).CCK-8 method was used to detect cell proliferation.The flow cytometry was used to detect cell apoptosis.Real-time fluorescence quantitative polymerase chain reaction(RT-qPCR),Western blot were used to detect the expression levels of B-cell lymphoma factor 2(Bel-2),Bel-2 associated X protein(Bax),and phosphatidylinositol 3-kinase(PI3K)mRNA and protein.Results Compared with those in the normal control group,the cell survival rate of H1299 cells significantly decreased(P<0.01),while the cell apoptosis rate significantly increased in the high-dose and low-dose GPM groups(P<0.01).The expression levels of PI3K and Bax mRNA and protein significantly decreased(P<0.01),while the expression levels of Bel-2 mRNA and protein significantly increased in the high-dose and low-dose GPM groups(P<0.05).Conclusion GPM can inhibit the proliferation of H1299 cells,and its mechanism may be related to the regulation of the PI3K,Bax,Bcl-2 mRNA and protein expression.
