没食子水提物对人牙周膜成纤维细胞生物学活性的影响

目的 探讨没食子水提物在防治口腔炎性疾病中的作用。方法 分离培养人牙周膜成纤维细胞(HPDLF),设立阴性对照组(PBS)、脂多糖组(1µg/mL)、没食子组(10 mg/mL)、氯己定组(0。2%,m/m)。对没食子水提物粉末进行大体检查,采用扫描电子显微镜观察没食子水提物粉末的微观特征。采用MTT法检测细胞活力,XTT法检测细胞增殖水平,以划痕伤口愈合迁移试验考察细胞迁移情况,采用酶联免疫吸试验(ELISA)法检测白细胞介素 1β(IL-1β)、白细胞介素 6(IL-6)和肿瘤坏死因子-α(TNF-α)表达水平。结果 没食子水提物粉末大体检查及显微特征明显。没食子组细胞活力显著高于其他组(P<0。05)。给药后第 3,5,7 天,没食子组HPDLF增殖水平显著高于其他组(P<0。05)。与阴性对照组比较,没食子组IL-1β、IL-6、TNF-α的表达水平显著降低(P<0。05)。没食子组细胞 24h划痕伤口几乎完全闭合。结论 没食子水提物可减轻HPDLF相关炎性反应。
Effect of Water Extract of Galla Turcica on the Biological Activity of Human Periodontal Ligament Fibroblasts
Objective To investigate the role of water extract of Galla Turcica in the prevention and treatment of oral inflammatory diseases.Methods The human periodontal ligament fibroblasts(HPDLFs)were isolated and cultured,and divided into the negative control group(PBS),the lipopoly saccharide group(1 µg/mL),the Galla Turcica group(10 mg/mL),and chlorhexidine group(0.2%,m/m).The gross examination was performed on the water extract powder of Galla Turcica,the scanning electron microscope(SEM)was used to observe the microscopic characteristics of the the above water extract powder.The MTT assay was used to measure cell viability,the XTT assay was used to detect cell proliferation level,the wound healing assay was used to investigate cell migration,and the enzyme-linked immunosorbent assay(ELISA)was used to detect the expression levels of interleukin-1β(IL-1β),interleukin-6(IL-6),and tumor necrosis factor-α(TNF-α).Results The gross examination and characteristics microscopic characteristics of water extract powder of Galla Turcica were obvious.The cell viability in the Galla Turcica group was significantly higher than that in the other groups(P<0.05).On the 3rd,5th,and 7th days after administration,the proliferation level of HPDLFs in the Galla Turcica group was significantly higher than that in the other groups(P<0.05).Compared with those in the negative control group,the expression levels of IL-1β,IL-6 and TNF-α in the Galla Turcica group significantly decreased(P<0.05).The wound healing assay after 24 h showed that the scratch wound in the Galla Turcica group almost completely healed.Conclusion The water extract of Galla Turcica can relieve the inflammatory reactions of HPDLFs.
