芩珠凉血合剂化学成分及抗银屑病活性研究

目的 筛选芩珠凉血合剂中具有治疗银屑病作用的化学成分。方法 采用色谱技术对芩珠凉血合剂进行分离纯化,经波谱数据鉴定化合物结构,采用MTT法进行体外抗银屑病活性研究。结果 从芩珠凉血合剂中分离鉴定出11种化合物,包括甘草素(化合物1),甘草苷(化合物2),芹糖甘草苷(化合物3),升麻素(化合物4),5-O-甲基维斯阿米醇苷(化合物5),黄芩苷(化合物6),黄芩素(化合物7),汉黄芩素(化合物8),丹皮酚(化合物9),苯甲酸(化合物10),对羟基苯甲酸乙酯(化合物11)。化合物2-3,5-7对角质形成细胞的增殖均有不同程度的抑制作用,化合物6对角质形成细胞的增殖抑制活性最强(半数抑制浓度为2。5 μmol/L)。结论 化合物1-11均为首次从芩珠凉血合剂中分离得到,具有抗角质形成细胞增殖的作用。
Chemical Compounds from Qinzhu Liangxue Mixture and Its Effect on Psoriasis
Objective To screen the chemical compounds in Qinzhu Liangxue Mixture that have therapeutic effects on psoriasis.Methods The compounds in the Qinzhu Liangxue Mixture were isolated and purified by the chromatography technology,the compound structures were identified by the spectral data,and the in vitro anti-psoriasis activity was studied by the MTT assay.Results Eleven compounds were isolated and identified from Qinzhu Liangxue Mixture,including liquiritigenin(compound 1),liquiritin(compound 2),liquiritin apioside(compound 3),cimifugin(compound 4),4'-O-β-D-glucosyl-5-O-methylvisamminol(compound 5),baicalin(compound 6),baicalein(compound 7),wogonin(compound 8),paeonol(compound 9),benzoic acid(compound 10),and ethylparaben(compound 11).Compounds 2-3 and 5-7 showed different degrees of inhibitory effects on the proliferation of keratinocytes,and compound 6 had the strongest inhibitory activity on the proliferation of keratinocytes,with a half maximal inhibitory concentration(IC50)of 2.5 μmol/L.Conclusion Compounds 1-11 were isolated for the first time from Qinzhu Liangxue Mixture and have anti-proliferative effects on keratinocytes.
