肠胃散高效液相色谱指纹图谱及化学模式识别研究

目的 为肠胃散的质量评价提供参考。方法 建立高效液相色谱(HPLC)指纹图谱,色谱柱为Agilent Eclipse XDB-C18柱(250 mm × 4。6 mm,5 μm),流动相为乙腈-0。2%磷酸溶液(梯度洗脱),流速为1。0mL/min,检测波长为226 nm,柱温为25 ℃,进样量为10 μL。采用中药色谱指纹图谱相似度评价系统(2012版)建立10批样品的HPLC叠加指纹图谱,并进行相似度评价。考察整体相似度和改良程度相似度方法的适用性,结合聚类分析法和主成分分析法综合评价其质量。结果 HPLC指纹图谱共识别出18个共有峰,确定峰1、峰7、峰9、峰11、峰14、峰16分别为橙皮苷、肉桂酸、桂皮醛、丁香酚、吴茱萸碱和吴茱萸次碱。指纹图谱评价系统相似度、整体相似度和改良程度相似度分别为0。996~1。000,0。886~0。955,0。878~0。951;按整体相似度和改良程度相似度分类,编号为S3-S4,S6-S7,S9的样品聚为一类,其余编号的样品聚为另一类。聚类分析法将10批样品聚为3类,编号为S3-S4,S6-S7,S9的样品,编号为S8,S10的样品,编号为S1-S2,S5的样品分别聚为一类。经主成分分析,4个主成分累积方差贡献率为90。662%,主成分综合得分排序按样品编号从大至小为S8>S9>S6>S3>S7>S10>S4>S5>S1>S2。结论 所建立的方法简单快速、可行性高,可用于全面评价肠胃散的质量。
HPLC Fingerprints and Chemical Pattern Recognition of Changwei Powder
Objective To provide a reference for the quality evaluation of Changwei Powder.Methods The high-performance liquid chromatography(HPLC)fingerprint was established,the chromatographic column was Agilent Eclipse XDB-C18 column(250 mm × 4.6 mm,5 µm),the mobile phase was acetonitrile-0.2%phosphoric acid solution(gradient elution),the flow rate was 1.0 mL/min,the detection wavelength was 226 nm,the column temperature was 25 ℃,and the injection volume was 10 μL.The Similarity Evaluation System for Fingerprint Chromatographic of Traditional Chinese Medicine(Version 2012)was used to establish HPLC overlay fingerprints of 10 batches of samples and conduct similarity evaluation.The applicability of methods for overall similarity and improved extent similarity was investigated,and the quality was comprehensively evaluated by combining clustering analysis and principal component analysis.Results A total of 18 common peaks were identified in the HPLC fingerprints,and peaks 1,7,9,11,14,and 16 were determined to be hesperidin,cinnamic acid,cinnamaldehyde,eugenol,evodiamine,and rutaecarpine,respectively.The similarity of fingerprints,overall similarity,and improved extent similarity were 0.996-1.000,0.886-0.955,and 0.878-0.951,respectively.According to the overall similarity and improved extent similarity,samples numbered S3-S4,S6-S7,and S9 were grouped into one category,while samples with other numbers were grouped into another category.Cluster analysis was used to group 10 batches of samples into three categories:samples numbered S3-S4,S6-S7 and S9,samples numbered S8 and S10,and samples numbered S1-S2 and S5.According to principal component analysis,the cumulative variance contribution rate of the four principal components was 90.662%.The comprehensive scores of the principal components were ranked in descending order of sample number as S8>S9>S6>S3>S7>S10>S4>S5>S1>S2.Conclusion The established method is simple,fast,and highly feasible,which can be used to comprehensively evaluate the quality of Changwei Powder.
