Simultaneous Determination of Six Components in Jinyinhua Oral Liquid by HPLC
Objective To establish a high-performance liquid chromatography(HPLC)method for simultaneous determination of neochlorogenic acid,chlorogenic acid,caffeic acid,rutin,isochlorogenic acid B,and isochlorogenic acid C in Jinyinhua Oral Liquid.Methods The chromatographic column was Shimadzu-GL C18 column(250 mm × 4.6 mm,5 µm),the mobile phase was acetonitrile-0.1%phosphoric acid aqueous solution(gradient elution),the flow rate was 1.0 mL/min,the detection wavelength was 320 nm,the column temperature was 30 ℃,and the injection volume was 10 μL.Results The linear ranges of neochlorogenic acid,chlorogenic acid,caffeic acid,rutin,isochlorogenic acid B,and isochlorogenic acid C were 6.52-62.5 μg/mL,15.48-154.8 μg/mL,1.82-18.2 μg/mL,2.62-26.2 μg/mL,3.34-33.4 μg/mL,2.18-21.8 μg/mL(R2 ≥ 0.999 3,n=6),respectively.The RSDs of precision,stability,and repeatability test results were all lower than 3.0%(n=6).The average recoveries of neochlorogenic acid,chlorogenic acid,caffeic acid,rutin,isochlorogenic acid B,and isochlorogenic acid C were 98.95%,101.37%,99.34%,97.85%,100.94%,and 98.95%,with RSDs of 2.06%,1.91%,2.54%,1.58%,2.18%,and 1.66%(n=6),respectively.The contents of six components in three batches of samples were in the ranges of 0.350 2-0.394 4 mg/mL,0.851 3-0.921 4 mg/mL,0.034 9-0.037 0 mg/mL,0.083 7-0.109 1 mg/mL,0.107 9-0.117 2 mg/mL,and 0.126 4-0.136 2 mg/mL,respectively.Conclusion This method is simple,accurate,with good specificity and repeatability,which can be used for the simultaneous determination of six components in Jinyinhua Oral Liquid.
