Antitumor Effect of Embelin on MCF-7 Breast Cancer Xenograft in Nude Mice
Objective To investigate the antitumor effect of embelin on the MCF-7 breast cancer xenograft in nude mice.Methods Thirty female Balb/c-nu nude mice were selected and randomly divided into the model group[equal volume of 0.025%ammonia solution(pH 8.2)],the 5-fluorouracil group(20 mg/kg),the embelin high-,medium-,and low-dose groups(400,200,100 μg/kg),with six mice in each group.The MCF-7 cell suspension(0.1 mL)was subcutaneously inoculated into the bilateral armpits of nude mice to construct the breast cancer xenograft in nude mice.After successful modeling,the nude mice in each group were given corresponding drugs or an equal volume of 0.025%ammonia solution(pH 8.2)by gavage or intraperitoneal injection once a day for 14 d.After the last administration,the tumor mass was weighed,and the tumor inhibition rate was calculated;the hematoxylin-eosin(HE)staining was used to observe tumor cell morphology,the Tunel staining was used to analyze tumor cell apoptosis and calculate apoptosis rate;the enzyme-linked immunosorbent assay(ELISA)was used to detect the tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6)levels in serum;the immunohistochemical staining was used to detect the expression level of Caspase-3 protein.Results Compared with those in the model group,the tumor mass in the 5-fluorouracil group and embelin high-dose group significantly decreased(P<0.01 or P<0.05);the apoptosis rate in each drug group significantly increased;the serum TNF-α level in the 5-fluorouracil group and embelin high-and medium-dose groups significantly increased(P<0.01 or P<0.05);the expression level of Caspase-3 protein in each drug group significantly increased(P<0.01).Conclusion Embelin can promote the apoptosis of MCF-7 cells in nude mice,and its mechanism may be related to the upregulation of Caspase-3 protein expression.
