Effect of Mahuang Decoction on the Immune Barrier Function of Nasal Mucosa in Model Rats
Objective To investigate the effect of Mahuang Decoction on the nasal mucosal barrier function in model rats and relevant mechanism.Methods Rat models of nasal mucosal immune dysfunction were constructed by cold-hot cycle stimulation and were given Mahuang Decoction by gavage.The nasal mucosa was taken for transcriptome sequencing,and the qPCR and Western blot were used for verification test.Results Mahuang Decoction could affect the β-interferon level,extracellular matrix,response to cytokines and other processes in model rats,and it also played a role by affecting the tumor necrosis factor signaling pathway,JAK-STAT signaling pathway,etc.Mahuang Decoction upregulated the expression of differential genes including LCN2,S100A8,MUC5b,MMP13,CXCL10,CCL20,IL17RA and the protein expression of IL-22,STAT3,p-STAT3 and p-STAT1,and decreased the mRNA expression of TRAF6 by activating the IL-22/STAT3 pathway.Conclusion Mahuang Decoction can improve the local immune barrier function in the nasal cavity of model rats through the IL-22/STAT3 signaling pathway.
