基于超高效液相色谱磐安白术指纹图谱的建立及模式识别研究

目的:采用超高效液相色谱(UPLC)建立浙江磐安地区白术生品的指纹图谱,并结合相似度计算和化学模式识别方法评价白术质量.方法:采用Acquity UPLC·BEHC18色谱柱(2.1mm×50mm,1.7μm),甲醇-水为流动相进行梯度洗脱,流速0.25mL/min,柱温25℃,检测波长240ntm,进样量10μL.对19批磐安地区白术生品和14批市售白术进行测定,计算相似度,并进行聚类分析和主成分分析.结果:建立了白术的指纹图谱测定方法和磐安地区白术指纹图谱的共有模式,其中磐安地区白术药材相似度均在0.95以上,聚类分析和主成分分析结果与其一致.结论:建立的白术生品指纹图谱特征性强,结合模式识别技术可用于白术的质量评价,为白术药材的全面质量控制提供科学依据.
Study on the fingerprint and chemical pattern recognition method for Pan'an atractylodes phizome based on UPLC
Objective:Establishing the fingerprint of Pan'an atractylodes phizome with UPLC method to evaluate the quality through the similarity calculation and chemical pattern recognition.Methods:Separation was performed at 25℃ on an Acquity UPLC BEH C18 column (2.1mm×50mm,1.7μm).Gradient elution was performed with the mobile phase of methanol and water.The flowing rate was 0.25mL/min.The UV detective wavelength was set at 240rim.The sample size was 10μL.Total 19 batches of Pan'an atractylodes rhizome and 14 batches of atractylodes rhizome from any other areas were analyzed.The similarity was calculated,then the cluster analysis and principal component analysis were analyzed.Results:The UPLC fingerprint was developed and the reference chromatographic fingerprint of Pan'an atractylodes rhizome was established.The results of similarity calculation were more than 0.95,which were in accordance with the results of the cluster analysis and principal component analysis.Conclusion:The fingerprint of atractylodes phizome established has a high characteristic,which could be used in the quality evaluation of atractylodes phizome if combined with pattern recognition that would provide a reference for the quality control of atractylodes phizome.
