基于RNA-seq技术的香连外洗液逆转白念珠菌耐药基因组学研究

摘要
目的:对香连外洗液诱导下恢复氟康唑敏感株及自然转种恢复敏感株差异基因进行转录组学研究.方法:通过测定最低抑菌浓度值(MIC)观察香连外洗液诱导及自然转种白念珠菌耐药株恢复药物敏感性所需的代数;对两种情况下恢复敏感的白念珠菌分别进行转录组测序;GO方法分析比较二者的差异基因.结果:香连外洗液作用下的耐药株转种至第6代恢复药物敏感性,后者转种至第16代恢复敏感性.转录组测序表明,与自然转种株相比,香连外洗液诱导株有174基因RPKM指标上调表达,164基因RPKM指标下调表达.结论:香连外洗液可诱导耐药株较早恢复药物敏感性;在其作用下,菌株蛋白质合成运输活动活跃,线粒体活动增强,应激性明显.
Abstract
Objective:To investigate genomics of restoration sensitivity of drug-resistant candida albicans strains to fluconazole induced by Xianglian Lotion and subcultivation.Broth microdilution test was used to detect minimal inhibitory concentration (MIC) of drug-resistant candida albicans strains induced by XiangLian Lotion and subcultivation.Transcriptome sequencing was used to determine and compare the transcript of sensitivity strains induced by Xianglian Lotion and subcultivation,and the differential genes were compared by GO method.Results:Xianglian Lotion could induce drug-resistent strains to restore sensitivity significantly in the 6th generations.Subcultivation was in the 16th generations.Transcriptome sequencing showed that sensitivity strains induced by Xianglian Lotion had 174 genes RPKM index upregulated,164 genes downregulated compared to subcultivation strains.Conclusion:Xianglian Lotion can induce drug-resistent strains to restore sensitivity early.Under the action of Xianglian Lotion,the protein synthesis of strain antimates and activity of mitochondrial are enhanced,the irritability is obvious.
