瓜蒌皮通过金属基质蛋白酶9促进高血脂合并心肌缺血大鼠CD34阳性细胞动员

目的:研究瓜蒌皮对高血脂合并心肌缺血(HL-AMI)大鼠CD334+细胞的动员作用,以及对金属基质蛋白酶9(MMP-9)蛋白表达量和酶活性的调节作用,从促进干细胞动员的角度揭示该药材干预冠心病痰浊壅塞证的机制.方法:雄性Wistar大鼠随机分为对照组、模型组、瓜蒌皮干预组,每组再分为7个亚组.各亚组大鼠分别在急性心肌缺血(AMI)后第1-7天处死,腹主动脉取血及心脏,检测外周血CD菇细胞含量及血浆、心肌组织中MMP-9含量.结果:AMI后,模型组大鼠CD34+细胞含量于第6天达到峰值并开始下降(P<0.05),MMP-9活性及表达量第3天达到峰值,第4天即下降到基线水平;瓜蒌皮干预组大鼠CD34+细胞含量7d内一直上升,且上升幅度明显高于模型组(P<0.05),MMP-9活性及表达量于第5天达到峰值,且一直明显高于同时期的模型组(P<0.05).结论:瓜蒌皮可显著促进HL-AMI大鼠骨髓干细胞的动员,其机制可能与上调MMP-9活性及蛋白表达量有关.
Trichosanthes pericarpium enhances the mobilization of CD34+ cells via matrix metalloproteinase 9 in the rat model of hyperlipemia complicated with acute myocardial ischemic
Objective:To investigate the effect of the Chinese herbal medicine trichosanthes pericarpium on the mobilization of CD34+ cells in hyperlipemia complicated with acute myocardial ischemia (HL-AMI) rats,and to reveal the mechanism in the perspective of regulating marrow stem cells mobilization.Methods:Wistar male rats were randomly divided into the control group,model group and the trich group,moreover,rats in each group were randomly redivided into 7 subgroups.Rats in the 1st-7th subgroup were phlebotomized on the lst-7th day after AMI model established respectively.CD34+ cells and the level of matrix metalloproteinase 9 (MMP-9) in the serum and in the myocardial tissue of each rat were determined.Results:During 7 days after AMI operation,in rats of the model group,the number of CD34+ cells increased rapidly during the 1st-6th day,reached its peak value and decreased obviously on the 6th day (P<0.05);the serum level of MMP-9 increased rapidly since the 3rd day,and decreased obviously since the 4th day (P<0.01).However,within the same phase,in the rats of the trich group,the blood contents of CD34+ cells and MMP-9 always increased,and were higher than those levels in the rats of model group during the whole experiment phase (P<0.05).Conclusion:In AMI rats,the trichosanthes pericarpium water decoction could significantly promote the mobilization of CD34+ cells and notably upregulate the expression of MMP-9,and the mechanism may be related to upregulates MMP-9 activity and protein expression
