Effects of Gegen Qinlian Decoction on methylation of Socs3 gene in adipose tissue from T2DM rats and correlations between methylation and physiological and biochemical parameters
Objective:To explore the effects of Gegen Qinlian Decoction(GQD)on methylation of Socs3 gene in the adipose tissue of rats with type 2 diabetes mellitus(T2DM)induced by streptozotocin and high-fat diet as well as the correlations between methylation and physiological and biochemical indicators.Methods:Forty rats were divided into four groups,namely,the control group,the model group,low-(5.5 g/kg),and high-dose(11.0 g/kg)GQD(GQDL and GQDH)groups,10 rats in each group.The epididymal adipose tissue of rats was collected for detecting mRNA level and all the cytosine methylation levels.Spearman correlation coefficient was used to analyze the correlations between C methylation in the amplified fragment and physiological and biochemical indicators.Results:GQDL and GQDH remarkably reversed the down-regulated mRNA level of Socs3 triggered by STZ and HFD(P<0.01).The higher percent methylation at CG3[part of the binding site of GATA binding protein 1(GATA1)]was found in model group compared to control group(P<0.05).Furthermore,methylation level of CG3 had significant positive correlations with glycosylated serum protein(GSP)(ρ=0.413,P=0.045)and inverse correlation with total cholesterol(TC)and high density lipoprotein cholesterol(HDL-c)(ρ=-0.432,P=0.035).However,the methylation of CG3 showed no significant difference between Model and GQDL or GQDH.Conclusion:GQD reverses effectively the expression of Socs3 in epididymal adipose tissue of T2DM rats.CG3 methylation near the promoter in Socs3 is associated with GSP,TC,and HDL-c,but this site may not be a target for GQD.
