Effects of Gengxin Tang on serum estrogen levels and hypothalamic estrogen receptors in a perimenopausal insomnia model in rat
Objective:To explore the effects of professor XIAO Chengjun,a renowned Chinese medicine practitioner,on the behavior,serum estrogen levels,and hypothalamic estrogen receptor expression of perimenopausal insomnia model rats using the clinical formula'Gengxin Tang'.Methods:Sixty 7-week-old female SD rats were selected,and a perimenopausal insomnia rat model was induced by gavage of Tripterygium wilfordii glycoside solution combined with intraperitoneal injection of p-chlorophenylalanine(PCPA)suspension.Twenty-six successfully modeled rats were randomly divided into a model group and a traditional Chinese medicine group,with 13 rats in each group,and an additional 12 rats in a blank group;The traditional Chinese medicine group was orally administered 1.8 g/kg of Gengxin Tang granules solution daily,while the blank group and model group rats were orally administered an equal amount of 0.9%sodium chloride solution daily for 14 days.Observing behavioral changes in rats using open field experiments(OFT),detecting serum estradiol(E2)levels using radioimmunoassay,detecting estrogen receptor(ER)protein expression in the hypothalamus of rats using Western Blot,and detecting ER in the hypothalamus of rats using RT-qPCR α,β mRNA expression.Results:Compared with the blank group,the OFT level and vertical activity score of the model group rats were significantly reduced(P<0.05),the number of modifications was significantly increased(P<0.01),serum E2 was significantly decreased(P<0.01),and hypothalamic ER was significantly reduced α,ER βsignificant decrease in protein expression(P<0.05),ER α/ER β ratio significantly increased(P<0.01);Compared with the model group,the modification frequency of the Chinese medicine group rats was significantly decreased(P<0.01),and the hypothalamic ER decreased β significant increase in protein expression(P<0.05),ER α/ER β ratio decreased(P<0.01).Conclusion:Gengxin Tang can upregulate the serum E2 level and the expression of different subtypes of ER proteins and genes in the hypothalamus of perimenopausal insomnia model rats,thereby improving the sleep status of rats.
