Discussion on the effect and mechanism of total saponins of Dioscoreae Nipponicae Rhizoma on hyperuricemia rats based on 16S rRNA sequencing
Objective:To investigate the effects of total saponins of Dioscoreae Nipponicae Rhizoma on intestinal flora in hyperuricemia rats.Methods:A total of 63 rats were randomly divided into control group(n=10)and model group(n=53).The model group was fed with 10%yeast feed and intragaxed with potassium oxyazinate solution for 14 days.Then the rats were randomly divided into model group,low-,middle-and high-dose of total saponins of Diascoreae Nipponicae Rhizoma group and allopurinol group,with 10 rats in each group.The rats were treated with allopurinol and total saponins of Dioscoreae Nipponicae Rhizoma from the 15th day and lasted for 2 weeks.The contents of serum uric acid,creatinine and urea nitrogen were analyzed by fully automated biochemical instrument,and the changes of intestinal flora were further detected by 16S rRNA sequencing.Results:All administration groups can significantly reduce uric acid level(P<0.01),and the high-dose of total saponins of Dioscoreae Nipponicae Rhizoma group can reduce urea nitrogen level(P<0.05),regulate the species abundance and evenness of intestinal flora,adjust the abundance of Proteobacteria,reduce the abundance of Bacteroidetes,and increase the abundance of Lactobacillus and Ruminococcaceae_Ruminococcus.Meanwhile,the abundance of Bacteroides and Parabacteroides was decreased.Conclusion:The total saponins of Dioscoreae Nipponicae Rhizoma can significantly reduce the level of uric acid in hyperuricemia rats,suggesting that the mechanism may be related to the regulation of intestinal flora disorder.
