Discussion on the effects of fried atractylodes-fried atractylodis on prevention and treatment of hyperlipidemia based on'spleen transporting and transforming water'
Objective:To construct a model for the prevention and treatment of hyperlipidemia,and to investigate the effects of fried atractylodes-fried atractylodis on aqueous metabolism in rats with hyperlipidemia.Methods:A total of 48 male SD rats were divided into blank control group,model control group,Ezetimibe group(1 mg/kg),fried atractylodes-fried atractylodis group(6 g/kg),fried atractylodes group(3 g/kg)and fried atractylodis group(3 g/kg),with 8 rats in each group.The blank control group was fed with ordinary diet,and the rest of the group was given high-fat diet for 6 consecutive weeks.Biochemical assays of blood lipid cholesterol(TC),triglyceride(TG),high-density lipoprotein cholesterol(HDL-C)and low-density lipoprotein cholesterol(LDL-C)in rats.ELISA was used to detect serum gastrin(GAS),motilin(MTL)and colonic Na+-K+-ATPase.PCR and Western Blot were used to detect colonic aquaporin(AQP)3 and AQP9 mRNA and protein.Results:Compared with the blank control group,the levels of TC,LDL-C,MTL,and AQP9 mRNA and protein in the model control group were significantly increased(P<0.05,P<0.01).The levels of HDL-C,GAS,Na+-K+-ATP were significantly decreased(P<0.01).Compared with the model control group,the levels of TC,TG,LDL-C,and the mRNA and protein levels of AQP9 in the fried atractylodes-fried atractylodis group were significantly decreased(P<0.05,P<0.01),the levels of GAS and Na+-K+-ATP were significantly increased(P<0.05,P<0.01).Conclusion:Fried atractylodes-fried atractylodis may be used to prevent hyperlipidemia by regulating Na+-K+-ATPase and AQPs-mediated water-liquid metabolism.
