Regulation of Mongolian medicine Tubson-2 Decoction on bone metabolism in zebrafish model with glucocorticoid-induced osteoporosis
Objective:To investigate the effects of Mongolian medicine Tabson-2 Decoction(TBD)and its characteristic components against glucocorticoid-induced osteoporosis of zebrafish.Methods:The zebrafish larva with 3 days post fertilization(3 dpf)were randomly divided into model group,positive control group,blank group,TBD administration group,administration groups with characteristic component monomers(3-hydroxycinnamic acid,astragaloside,chlorogenic acid,caffeic acid and protocatechuic acid)and compatibility.Glucocorticoid-induced osteoporosis model was induced by 25 g/mL prednisolone.The administration group was given low,medium and high concentrations of drugs,respectively.The larva were cultured to 7 dpf,stained with alizarin red S,observed and photographed under stereomicroscope,and the number of vertebrae and the stained area of spinal bone were calculated.Results:Prednisolone model group showed successful modeling;the number and staining area of zebrafish vertebral bodies in TBD group were significantly higher than those in model group(P<0.05,P<0.01),which showed a certain dose-response relationship;Monomer characteristic components and their compatibility can increase the number and staining area of vertebral bodies in varying degrees,compared with the model group,the number and staining area of vertebral bodies in the high concentration compatibility group were significantly increased(P<0.05).Conclusion:TBD can effectively regulate zebrafish glucocorticoid-induced osteoporosis,the therapeutic effect of the compatibility of characteristic components on osteoporosis is better than that of each monomer characteristic component.
