Effect and mechanism of alcohol extract from Forsythia suspensa leaves after frost on attenuating Aβ toxicity of model nematode of Alzheimer's disease
Objective:To explore the anti-amyloid β(Aβ)toxicity of alcohol extract of Forsythia suspense leaves after frost(AFLF)in Alzheimer's disease(AD)model nematode,and to discuss its mechanism.Methods:The effects of AFLF on lifespan,paralysis time,chemotaxis,Aβ deposition,reactive oxygen species and expression levels of oxidative stress-related genes(daf-16,skn-1,hsf-1,ctl-1,sod-3)in AD model nematodes were examined to elucidate the anti-Aβ toxicity of AFLF and its mechanism.Results:Compared with the control group,5 mg/mL AFLF could significantly prolong the life span of AD model nematode(P<0.01),reduce Aβ-induced paralytic toxicity and chemotactic defects,reduce Aβ deposition,and significantly increase the expression of daf-16,sod-3,hsf-1,skn-1 and ctl-1(P<0.01).Conclusion:AFLF can reduce Aβ-induced toxicity in AD model nematodes by reducing oxidative stress response and inhibiting Aβ-deposition.
