Study on the central mechanism of acupuncture at Taichong(LR 3)acupoint on regulation of SHRs blood pressure based on the renin-angiotensin system of medulla oblongata
Objective:To observe the regulative effect of acupuncture manipulation with definite antihypertensive effects on the renin-angiotensin system(RAS)in the medulla oblongata of spontaneously hypertensive rats(SHRs)for 28 days,further clarifying the central mechanism of its antihypertensive effects.Methods:A total of 30 male SHRs were randomly and equally divided into a model group,an acupuncture without manipulation group and an acupuncture manipulation group,and 10 WKY rats were assigned to the control group.The according acupuncture intervention at Taichong(LR 3)acupoint were administered in the two acupuncture groups,the rats in the blank group and model groups received no treatment.All interventions were carried out once a day,with a day off between two weeks,a total of 28 days.The blood pressure of rats was measured repeatedly,and the expression of RAS regulatory axis components in medulla oblongata was detected by ELISA and RT-qPCR on the 28th day of the experiment.Results:From the 7th day of acupuncture,the blood pressure of SHRs in both acupuncture groups was significantly decreased than that in mode group,the antihypertensive effects lasted until the 28th day of acupuncture(P<0.05,P<0.01),and the hypotensive effect was more obvious after applying acupuncture manipulation(P<0.01);Compared with model group,in the acupuncture manipulation group,the expression of ACE and Ang Ⅱ in the RAS elevating-BP axis were significantly decreased(P<0.05,P<0.01);at the same time,the expression of ACE2,Ang(1-7)and MasR in the RAS lowering-BP axis were significantly up-regulated(P<0.01,P<0.05).Conclusion:Acupuncture intervention on Taichong(LR 3)point has a definite antihypertensive effect,and the longer time the acupuncture intervention is,the greater the hypotensive amplitude is,moreover,acupuncture can benignantly regulate the expression of RAS components in the medulla oblongata,especially on the lowering-BP axis ACE2-Ang(l-7)-MasR,which may be one of the central mechanisms of acupuncture on stable antihypertensive effects.
