Effects of tanshinone ⅡA on endometrial interstitial cells fibrosis through TGF-β1/PI3K/Akt signaling pathway
Objective:To investigate the effect of tanshinone ⅡA(TanⅡA)on human endometrial interstitial cells strain(HESCs)fibrosis through TGF-β1/PI3K/Akt signaling pathway.Methods:HESCs were infected with 10 ng/mL TGF-β1 to induce fibrosis,and HESCs was divided into 5 groups:normal control group,model group,Tan ⅡA low,medium,high dose group.The cytotoxicity and proliferation of HESCs were detected by CCK-8.The fluorescence intensity of α-SMA and Col1 α1 was detected by immunofluorescence.The protein expressions of α-SMA,Col1 α1,PI3K,Akt and p-Akt were detected by Western Blot.Results:The concentration of Tan ⅡA in 10-40 μg/mL had no toxicity to HESCs.Compared with normal control group,cell proliferation capacity,fluorescence intensity of α-SMA and Col1 α1,protein expression of α-SMA,Col1 α1,PI3K and p-Akt were increased in model group(P<0.05,P<0.01).Compared with model group,the cell proliferation ability of Tan ⅡA high dose group was decreased(P<0.05).The fluorescence intensity of α-SMA and Col1 α1 and the protein expressions of α-SMA,PI3K,p-Akt and Col1 α1 decreased in each Tan ⅡA intervention group(P<0.01,P<0.05),but there were no significant differences in the expression of Akt among all groups.Conclusion:Tan ⅡA can effectively antiendometrial fibrosis by inhibiting the TGF-β1/PI3K/Akt signaling pathway.
