复方西羚解毒胶囊调控JAK2-STAT3信号通路抗H1N1病毒感染作用

目的:研究复方西羚解毒胶囊抗H1N1病毒感染作用,并探讨其初步作用机制.方法:建模给药后,解剖,测定小鼠肺指数,肺病毒载量,肺组织病理变化,血清中肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)、白介素-6(IL-6)细胞因子分泌水平,肺泡巨噬细胞分型及眼球血CD4+/CD8+T细胞比例,检测JAK2-STAT3信号通路相关基因mRNA和蛋白表达.结果:与模型组比较,复方西羚解毒胶囊高、中剂量组可显著降低小鼠肺指数(P<0.01),抑制H1N1病毒的复制载量(P<0.01,P<0.05),减轻肺组织病理状况,显著降低TNF-α、IL-1β、IL-6的分泌(P<0.05,P<0.01),显著升高肺泡巨噬细胞M2抗炎表型的比例(P<0.01),降低外周血CD4+/CD8+T细胞比例(P<0.01),降低IFN-β、JAK2、STAT3、OAS1 mRNA表达(P<0.01,P<0.05),降低p-JAK2/JAK2、p-STAT3/STAT3蛋白表达(P<0.01).结论:复方西羚解毒胶囊对H1N1病毒感染具有显著的治疗效果,这可能是通过调节JAK2-STAT3信号通路发挥炎性控制和免疫调节作用实现的.
Anti-H1N1 effect of Compound Xiling Jiedu Capsules via the JAK2-STAT3 signaling pathway
Objective:To study the anti-H1N1 virus effect of compound Xiling Jiedu Capsules and its primary mechanism.Methods:After modeling and administration,the lung index,lung viral load,pathological changes of lung tissue,serum levels of TNF-α,IL-1β,IL-6 cytokine secretion,alveolar macrophage typing and the proportion of CD4+/CD8+T cells in eye blood were determined by dissection.mRNA and protein expression of genes related to JAK2-STAT3 signaling pathway were detected.Results:Compared with the model group,compound Xiling Jiadu Capsules medium-and high-dose groups could reduce lung index(P<0.01),inhibit the replication of H1N1 virus(P<0.01,P<0.05),alleviate the pathological condition of lung tissue,reduce the secretion of TNF-α,IL-1β and IL-6(P<0.05,P<0.01),significantly increase the proportion of anti-inflammatory phenotype M2 in alveolar macrophages(P<0.01),suppress the proportion of CD4+/CD8+T cells in peripheral blood(P<0.01),decrease the mRNA expression of IFN-β,JAK2,STAT3 and OAS1(P<0.01,P<0.05),and inhibit the protein expression of p-JAK2/JAK2 and p-STAT3/STAT3(P<0.01).Conclusion:Compound Xiling Jiedu Capsules has significant therapeutic effect on H1N1 virus infection,which may be achieved by regulating JAK2-STAT3 signaling pathway to exert inflammatory control and immune regulation.
