Study on the mechanism of Saussurea Involucrata Oral Liquid affecting intestinal flora to improve joint inflammation and cytokine expression in arthritis rats
Objective:To determine whether Saussurea Involucrata can improve joint inflammation and cytokines by changing intestinal flora in rats.Methods:A rat model of type ⅠⅡ collagen-induced arthritis(CIA)was established,treated with Saussurea Involucrata Oral Liquid,treated again after intestinal flora failure,and transplanted intestinal flora(FMT),and the swelling degree of the paws of rats in each group was observed.ELISA was used to detect the inflammatory factors interleukin(IL)-1 β,IL-6,IL-10,and tumor necrosis factor-α(TNF-α)in each group,HE staining was used to observe synovial pathology,and 16SrRNA was used to detect intestinal flora.Results:Compared with the model group,IL-1 β and IL-6 in the joints of rats in the Saussurea Involucrata group and Tripterygium wilfordii group were decreased(P<0.05),IL-10 and TNF-α were increased(P<0.01),and joint swelling was reduced(P<0.01).The intestinal flora is closer to the normal group than the model group;after intestinal flora transplantation,the joint swelling of rats in the Saussurea Involucrata feces group,Tripterygium wilfordii feces group,normal feces group,and normal rat gavage Saussurea Involucrata feces group improved(P<0.01),and the synovial proliferation and inflammatory cell infiltration are reduced.After removal of the intestinal flora intervention,and after treatment with Saussurea Involucrata,there was no statistically significant difference in joint inflammation in rats in the Saussurea Involucrata flora depletion group compared with the saline flora depletion group.Conclusion:Saussurea Involucrata Oral Liquid can affect inflammatory factors and improve joint inflammation by changing intestinal flora.
