桂枝加龙骨牡蛎汤对魄不安于肺不寐大鼠焦虑样行为的镇静机制

目的:从方剂组成上考察桂枝加龙骨牡蛎汤对魄不安于肺不寐大鼠焦虑样行为的作用及其镇静机制.方法:将72只雄性SD大鼠随机分为正常组、模型组、右佐匹克隆组(西药组)、桂枝加龙骨牡蛎汤组(中药组)、中药组去龙骨牡蛎组、中药组去牡蛎组、中药组去龙骨组、龙骨牡蛎组,前4组每组12只,后4组每组6只(只进行活体功能检测).采用自制狭长多平台浅水环境法剥夺除正常组外的其他各组大鼠睡眠42 d.第15-42天进行相应干预后进行肺功能检测、高架十字迷宫实验和睡眠监测.以尼氏染色观察前4组扣带回神经细胞变化,以免疫组化法检测前4组脑扣带回γ-氨基丁酸A受体γ2(GABAARγ2)、氮甲基天冬氨酸受体(NMDAR)和谷氨酸代谢型受体1(mGluR1)蛋白的表达.结果:与正常组比较,模型组潮气量、每分通气量、吸气峰值流速、开放臂进入次数和停留时间、开放臂停留时间比例、NREM和REM、平均睡眠片段时间、GABAARγ2蛋白均显著降低(P<0.01,P<0.05);呼吸频率、觉醒次数、NMDAR和mGluR1蛋白表达均显著升高(P<0.01);神经元尼氏体变小,排列紊乱,胶质细胞增多.与模型组比较,西药组和中药组开放臂进入次数和停留时间、开放臂停留时间比例、REM、NREM、睡眠片段平均时间、GABAARγ2蛋白表达显著升高(P<0.01,P<0.05);呼吸频率、觉醒次数、NMDAR和mGluR1蛋白表达显著下降(P<0.01),神经元尼氏体较多;与西药组比较,中药组开放臂停留时间和比例、REM、mGluR1蛋白表达显著升高(P<0.01),神经元尼氏体增多明显,排列整齐,胶质细胞少.中药组去龙骨牡蛎组、中药组去牡蛎组、中药组去龙骨组、龙骨牡蛎组与模型组呼吸功能监测和高架十字迷宫实验结果比较均存在差异有统计学意义的指标,但指标改善相对弱于中药组.结论:桂枝加龙骨牡蛎汤对魄不安于肺不寐大鼠具有显著疗效,且该疗效与桂枝汤调和阴阳营卫和龙骨牡蛎相须安肺魄的协同作用有关.
Sedative mechanism of Guizhi Jia Longgu Muli Decoction on the anxiety-like behaviour of the insomnia model rats with Po unrest in the lung
Objective:To examine the sedative effect and mechanism of Guizhi Jia Longgu Muli Decoction on the anxiety-like behaviour of the insomnia model rats with Po unrest in the lung from the composition of the prescription.Methods:Seventy-two rats were randomly distributed into normal group,model group,Eszopiclone group,Guizhi Jia Longgu Muli Decoction group(each of the former 4 groups has 12 rats),Guizhi Decoction group,Guizhi Jia Longgu Decoction group,Guizhi Jia Muli Decoction group,Longgu Muli group(each of the later 4 groups has 6 rats and only their whole body functions were detected).The rats except the normal group were deprived of sleep for 42 days by self-made narrow multi-platforms in the shallow water environment.From the 15th day to the 42nd day,the rats were intervened correspondingly.Then pulmonary function examination,elevated plus maze test and EEG monitoring were performed.The cingulate gyrus nerve cells were observed by nissl staining,and the GAB AAR γ 2,NMDAR,mGluR1 proteins in the brain cingulate gyrus were detected by immunohistochemistry method in the first 4 groups.Results:Compared with the normal group,pulmonary TVb,MVb,PIF,the number of open arm entries and stay time,proportion of open arm stay time,NREM,REM,average time of each sleep bont and GABAAR γ 2 decreased(P<0.01,P<0.05),while the respiratory frequency,the times of arousal,NMDAR and mGluRl increased in the model group(P<0.01,P<0.05).Nissl bodies of neurons became smaller and arranged disorderly;glial cells increased.Compared with the model group,the number of open arm entries and stay time,proportion of open arm stay time,REM,NREM,average time of each sleep bout,GABAAR γ 2 protein increased significantly(P<0.01,P<0.05),respiratory frequency,the times of arousal,the expression of NMDAR and mGluR1 protein decreased significantly(P<0.01)in Eszopiclone group and Guizhi Jia Longgu Muli Decoction group with more nissl bodies.Compared with the Eszopiclone group,open arm residence time and ratio,REM,mGluRl protein expression were significantly increased in the Chinese medicine group(P<0.01),nissl bodies of neurons increased and arranged orderly,glial cells decreased.Compared with model group,there were some statistical differences in the indexes of respiratory monitor and high plus maze test in the Guizhi Decoction group,Guizhi Jia Longgu Decoction group,Guizhi Jia Muli Decoction group,Longgu Muli group,but their effects were weaker than that of the Guizhi Jia Longgu Muli group.Conclusion:The curative effect of Guizhi Jia Longgu Muli Decoction on the insomnia rats with unrest Po in the lung does exist and is related to the synergism yin-yang and ying-wei regulation of Guizhi Decoction and Po sedation of Longgu Muli.
