基于IL-6/JAK2/STAT3信号通路调控神经元凋亡研究左归丸改善产前应激孕鼠子代学习能力的作用及机制

目的:基于白细胞介素6(IL-6)/酪氨酸蛋白激酶2(JAK2)/转录激活因子3(STAT3)信号通路探讨左归丸改善产前应激(PS)孕鼠子代学习能力的作用机制.方法:孕鼠50只,随机分为空白组,模型组,抗IL-6组,左归丸低、高剂量组.除空白组外,其他组通过慢性不可预测轻度应激复制PS孕鼠模型.自孕12 d起,左归丸低、高剂量组分别给予左归丸0.39、0.78 mg/kg灌胃,空白组和模型组以10 mL/kg的0.9%氯化钠溶液灌胃,抗IL-6组以10 μg/d腹腔注射IL-6抗体,直至产仔.糖水偏好实验、旷场实验进行模型评价,Morris水迷宫法评估仔鼠学习能力,ELISA法检测母鼠及仔鼠血清IL-6含量,尼氏染色评估仔鼠大脑海马神经元成熟个数,TUNEL法荧光检测海马神经元凋亡水平,Western Blot法检测仔鼠大脑海马IL-6和JAK2、STAT3蛋白及其磷酸化的表达水平.结果:与空白组比较,模型组仔鼠学习能力下降(P<0.01),海马成熟神经元数量减少及凋亡增加(P<0.01),母鼠及仔鼠血清IL-6含量增高(P<0.01),仔鼠海马IL-6蛋白及JAK2、STAT3磷酸化蛋白表达增高(P<0.01);与模型组比较,左归丸各剂量组和抗IL-6组不同程度的改善学习能力(P<0.01);仔鼠海马神经元成熟个数增加及凋亡减少(P<0.01,P<0.05);母鼠、仔鼠血清中IL-6以及仔鼠大脑海马IL-6蛋白及JAK2、STAT3磷酸化蛋白表达下调(P<0.01).结论:左归丸孕期干预PS孕鼠影响其子代学习能力的机制可能是通过IL-6/JAK2/STAT3信号通路介导的神经元凋亡.
Effects of Zuogui Pills on the learning ability of offspring of prenatal stress in pregnant rats through the IL-6/JAK2/STAT3 signaling pathway
Objective:To explore the mechanism of Zuogui Pills in protecting learning ability of offspring under prenatal stress(PS)based on IL-6/JAK2/STAT3 signaling pathway.Methods:A total of 50 pregnant rats were randomly divided into control,model,anti-IL-6 and Zuogui Pills low and high dose groups.PS pregnant rat models were replicated by chronic unpredictable mild stress(CUMS)during the whole pregnancy in all groups except the blank group.From the 12th day of pregnancy,the intragastric doses of Zuogui Pills in the low-dose group and high-dose group were 0.39 and 0.78 mg/kg,respectively.The normal group and model group were intragastrically administered with 10 mL/kg normal saline.The dams in the anti-IL-6 group were intraperitoneally injected with IL-6 antibody 10 pg/day until the rats give birth.The PS model was evaluated through the sugar water preference experiment and the open field test.The learning and memory abilities of offspring assessed by Morris water maze.The serum interleukin-6(IL-6)contents of the dams and the litter by enzyme-linked immunosorbent assay(ELISA).The number of immature neurons in the hippocampus of the offspring was assessed by Nissl staining.Terminal deoxynucleotidyl transferase dUTP nick end labeling(TUNEL)method was used to detect the apoptosis level of hippocampal neurons by fluorescence,the expression levels of IL-6,tyrosine protein kinase 2(JAK2)and transcriptional activator 3(STAT3)proteins with their phosphorylated proteins in hippocampus of the rats were detected by Western Blot.Results:Compared with the blank group,the learning ability of the offspring in the model group decreased(P<0.01);the number of mature hippocampal neurons decreased and apoptosis increased(P<0.01);the serum IL-6 content of the dams and offspring increased(P<0.01);the expression of IL-6 protein and JAK2 and STAT3 phosphorylated proteins in the hippocampus of offspring mice increased(P<0.01).Compared with the model group,each dose group of Zuogui Pills and the anti-IL-6 group improved learning ability to varying degrees(P<0.01);the number of mature hippocampal neurons in offspring increased and apoptosis decreased(P<0.01,P<0.05);the expression of IL-6 in the serum of dams and offspring,as well as IL-6 protein and JAK2 and STAT3 phosphorylated proteins in the brain hippocampus of offspring were down-regulated(P<0.01).Conclusion:The mechanism of PS mothers affecting their offspring learning ability may be neuronal apoptosis mediated by IL-6/JAK 2/STAT3.
