Simultaneous determination of emodin and chrysophanol in Baibi Sanhuang Ointment by HPLC
Objective:To develop a high performance liquid chromatography(HPLC)method for simultaneous determination of emodin and chrysophanol in Baibi Sanhuang Ointment.Methods:Isocratic elution was performed on a RD-C18 column(4.6 mm×250 mm,5 μm)with methanol-0.1%phosphoric acid solution(80:20)as mobile phase,the detection wavelength was set at 254 nm,the column temperature was 30 ℃,and the flow rate was 1.0 mUmin.Results:The separation degree of emodin and chrysophanol were good,their linear range were 0.19-6.04 μg/mL(r=0.9999),1.00-31.88 μg/mL(r=1.0000),respectively.The RSDs of precision,repeatability,and stability were ≤1.86%;and the average recovery were in the range of 96.8-103.6(RSD≤3.7%);The average content ranges of emodin and chrysophanol in 3 batches of Baibi Sanhuang Ointment were 16.78-22.51 and 117.19-131.65 μg/mL,respectively.Conclusion:The method developed in this study was accurate,and the repeatability was good,which can quickly achieve determination of emodin and chrysophanol in Baibi Sanhuang Ointment,and to provide reference for the quality control of Baibi Sanhuang Ointment.
