Protective effects of Zhonghua Dieda Pills on rats with acute soft tissue injury
Objective:To investigate the protective effect and mechanism of Zhonghua Dieda Pills on rats with acute soft tissue injury.Methods:A model of acute soft tissue injury in rats was established by using the vertical fall of heavy objects from high altitudes to cause soft tissue injuries.A total of 180 rats were randomly divided into normal control group,model group,positive drug group,and high,medium and low(2,1,0.5 g/kg)dose groups of Zhonghua Dieda Pills,with 30 rats in each group.On the 1st,3rd and 5th days of giving the corresponding drugs by gavage,10 rats were randomly sacrificed in each group,and the swelling degree,tenderness threshold,tumor necrosis factor α(TNF-α),interleukin 1β(IL-1β),prostaglandin E2(PGE2)and nerve growth factor(NGF)in local tissues were measured,and the pathology of muscle tissue was observed and scored by HE staining.Results:Compared with the model group,each dose group of Zhonghua Dieda Pills could significantly increase the tenderness threshold(on the 1st,3rd and 5th days of administration)(P<0.05,P<0.01),reduce the local swelling and TNF-α,IL-1β and PGE2,increase the NGF(on the 3rd and 5th days of administration)(P<0.01,P<0.05),and reduce the total score of muscle lesions(on the 5th day of administration)(P<0.05).The medium-and high-dose groups of Zhonghua Dieda Pills could significantly reduce the total score of muscle lesions(on the third day of administration)(P<0.05).The high-dose group of Zhonghua Dieda Pills could also significantly inhibit PEG2 level,reduce the total score of muscle lesions,and increase the content of NGF(on the first day of administration)(P<0.05,P<0.01).Conclusion:Zhonghua Dieda Pills have obvious protective effect on the acute soft tissue injury of rat model,and their mechanism is related to reducing the levels of TNF-α,IL-1 β,PGE2 and increasing the content of NGF in the injured area.
