Effects of Xinshubao Tablets on cardiac function and gelsolin in mice after myocardial infarction
Objective:To explore the protective effect of Xinshubao Tablets on cardiac function in mice after myocardial infarction(MI)and its influence on gelsolin(GSN).Methods:MI model was established by ligating LAD in mice.The C57BL/6J male rats were divided into sham group,model group,low dose of Xinshubao Tablets(XSB-L)group,high dose of Xinshubao Tablets(XSB-H)group,and Captopril(CAP)group,5 in each group.XSB-L group was given Xinshubao Tablets suspension(0.26 g·kg-1 d-1),XSB-H group was given Xinshubao Tablets suspension(0.52 g·kg-1 d-1),CAP group was given Captopril suspension(4.875 mg·kg-1·d-1),sham group and model group were given double distilled water.And they were given corresponding liquid intragastric treatment for 4 weeks.The heart function of the rats was detected by ultrasound,the specific cell types and genes impacted by MI was utilized to identify by single-cell sequencing technology.The expression of GSN in myocardial tissue was detected by Western Blot.Results:Compared with the model group,the cardiac index was significantly reduced(P<0.05);LVEF increased significantly(P<0.05).Single cell sequencing of the model group analysis demonstrated the enrichment of Ryr2,Myh6,Atp2a2,Actc1,Myl3,and Tnnt2 in the myocardial cells,Fibroblasts show increased GSN,COL1a1 and COL3a1,Endothelial cells show enriched PECAM1,EGFL7 and VEGFA.KEGG involved cardiac muscle contraction,hypertrophic cardiomyopathy,dilated cardiomyopathy,etc.Western Blot also showed that the expression of GSN decreased significantly in XSB_H group(P<0.05).Conclusion:The intervention of XSB can inhibit the expression of GSN protein in myocardial tissue of mice after MI,reduce fibrosis of MI tissue,reverse left ventricular remodeling,improve left ventricular function and improve the quality of life of mice after MI.
