Study on the protective effect and mechanism of the classic formula Xiaochengqi Decoction on gastric mucosa
Objective:To explore the protective effect of Xiaochengqi Decoction(XCQD)on injured gastric mucosa in vitro and in vivo,and elucidate the potential mechanism.Methods:HPLC was used to define the components in XCQD.MTT method and AnnexinV-PI double staining method were used to evaluate the protective effect of XCQD on oxidative damage cells in vitro.To evaluate the antioxidant effect of XCQD in vitro,the contents of SOD,GSH,and MDA were determined.In vivo study,SD rats were randomly divided into five groups:normal group,model group,low dose treatment group,high dose treatment group,and positive drug group.Histological analysis,analysis of inflammatory and oxidative factors in serum,and Western Blot were used to investigate the potential mechanism of XCQD on protecting acute gastric mucosal injury in vivo.Results:XCQD promotes cell proliferation,with the best effect observed at 0.7 mg/mL,reduces ROS,MDA,GSH in GES-1 cells,increases SOD levels to alleviate the damage of GES-1 cells induced by H2O2,and alleviates damage to gastric mucosal in rats caused by anhydrous ethanol,with an inhibition rate of 94.32%.In addition,XCQD also upregulated the expression of ZO-1 and Occludin and inhibited the NF-κB signaling pathway.Conclusion:XCQD can promote cell proliferation and reduce apoptosis by reducing oxidative stress and inflammatory response,inhibiting NF-κB signaling pathway and enhancing the tight junction structure,thereby exerting a protective effect on the gastric mucosa.
