Research on material basis of lotus plumule for anti-tumor from medicine-hebology perspective
Objective:To study the material basis of the anti-tumor efficacy of lotus plumule from the perspective of medicine-hebology.Methods:The particle size and morphology of neferine/manganese ion/human serum albumin composite nanoparticle was characterized by dynamic light scattering(DLS)and transmission electron microscope(TEM).The toxicity and ability to generate reactive oxygen species of neferine/manganese ion/human serum albumin composite nanoparticle on 4T1 cells was evaluated by CCK-8 assay and reactive oxygen species assay kit,respectively.The lipopolysaccharide,interleukin-4,neferine,manganese ion and neferine/manganese ion/human serum albumin composite nanoparticle were co-incubated with RAW264.7 cells for 24 h each and the ability of the neferine/manganese ion/human serum albumin composite nanoparticle to reprogramme macrophages was evaluated by detecting the percentage of M1 and M2 type macrophages using flow cytometry.4T1 tumor-bearing Balb/c mouse model was used to further evaluate the antitumor ability of neferine/manganese ion/human serum albumin composite nanoparticles in vivo.Results:In ultrapure water,the hydrodynamic diameter of neferine/manganese ion/human serum albumin composite nanoparticle was about 148 nm,The result of TEM showed that the neferine/manganese ion/human serum albumin composite nanoparticle was rounded and uniformly distributed.The CCK-8 experiments showed that there was a concentration-dependent cell killing of 4T1 cells by neferine/manganese ion/human serum albumin composite nanoparticle.Further mechanistic findings showed that it increased intracellular reactive oxygen species levels in 4T1 cells and effectively inhibited M2-type macrophage polarisation compared to the free neferine group.In vivo anti-tumor experiments confirmed that the anti-tumor efficacy of neferine/manganese ion/human serum albumin composite nanoparticles was stronger than that of neferine(P<0.05).Conclusion:The neferine/manganese ion/human serum albumin composite nanoparticle is found to be the novel anti-tumour pharmacodynamic substances of lotus plumule.
