酸枣DNA条形码筛选、种质资源鉴定与遗传多样性分析

摘要
目的:筛选特异性DNA条形码,并利用DNA条形码对酸枣不同种质进行种质资源鉴定及遗传多样性分析.方法:利用PCR扩增获得不同种质酸枣的ITS2、psbA-trnH、matK和rbcL核酸序列.用Mega和DNAman软件进行多重比对分析,筛选出的理想DNA条形码序列进行遗传距离分析,用Kimura 2-Parameter(K2P)模型分析遗传距离,用邻接法(NJ)构建系统发育树.结果:ITS2,psbA-trnH,matK和rbcL的扩增、测序成功率均为100%.所得序列与NCBI数据库上酸枣序列进行对比,鉴定率均>99%.其中psbA-trnH序列平均GC含量最低为31.9%,22个酸枣栽培种质的psbA-trnH序列一致率最低,psbA-trnH序列可能提供更多的变异信息.基于psbA-trnH序列构建的NJ系统发育树将酸枣种质分为两大类,自展支持率为100%.结论:psbA-trnH序列在酸枣种质中遗传多样性更高,更适用于酸枣不同种质亲缘关系研究.
Abstract
Objective:The specific DNA barcodes were screened,and the DNA barcodes were used to identify the germplasm resources and analyzed the genetic diversity of Ziziphus jujuba var.spinosa.Methods:The nucleic acid sequences of ITS2,psbA-trnH,matK and rbcL were amplified by PCR.Mega and DNAman software were used for multiple comparison analysis,K2P model was used to analyze genetic distance,and adjacency method(NJ)was used to construct phylogenetic tree.Results:The amplification and sequencing success rates of ITS2,psbA-trnH,matK and rbcL were 100%.The obtained sequence was compared with the sequence on the NCBI database,and the identification rate was greater than 99%.The average GC content of psbA sequence is 31.9%,the psbA-tmH sequence of 22 varieties of Z jujuba var.spinosa has the lowest consistency,and the psbA-trnH sequence may provide more variation information.The psbA-trnH sequence of new varieties of Z.jujuba var.spinosa were different.The NJ phylogenetic tree constructed based on the psbA-trnH sequence divides jujube germplasm into two categories,with a self expanding support rate of 100%.Conclusion:The psbA-tmH sequence has higher genetic diversity in jujube germplasm,making it more suitable for studying the genetic relationships among different jujube germplasm.
