Effects of Taohong Yin in atherosclerosis lymphangiogenesis based on the vascular endothelial growth factor C pathway
Objective:To explore the mechanism of Taohong Yin regulating lymphatic hyperplasia and improving atherosclerosis(AS).Methods:APOE-/-mice were divided into the model group,Taohong decoction low,medium,and high dose group,atorvastatin group,model+Vascular endothelial growth factor C(VEGFC)group,and Taohong decoction low,medium,and high dose+VEGFC group.The APOE-/-mice were induced by a high-fat diet and intervention with drugs and adeno-associated viruses.The mice were treated with a regular diet,normal saline,and control virus.The mice's body weight and blood lipids were detected,and the phenotype of AS mice was evaluated by pathological staining of aortic sinus and aortic arch.Primary bone marrow macrophages were extracted from tibia and femur of mice in each group,and their lipid load was detected.The expression of VEGFC pathway related molecules was detected by Western Blot and immunofluorescence.Results:Taohong Yin could reduce the body weight and serum lipid level of AS mice,reduce the area of aortic sinus plaque,the lipid content of aortic sinus and aortic arch,increase collagen content to stabilize the plaque,inhibit inflammatory infiltration and lymphangiogenesis,reduce the lipid load of primary bone marrow macrophages,and improve the tube formation of lymphatic endothelial cells induced by primary bone marrow macrophages.Taohong Yin could reduce the protein levels of VEGFC,FLT-4,LYVE-1,Prox-1,MMP9,and IL-1 βin AS mice and primary bone marrow macrophages.Conclusion:Taohong Yin can effectively improve AS,and its mechanism may be related to the inhibition of VEGFC/FLT-4 pathway to reduce lymphangiogenesis.
