健脾止动汤对慢性束缚应激多发性抽动症大鼠糖代谢影响及机制研究

目的:探究健脾止动汤对慢性束缚应激多发性抽动症(TS)大鼠模型糖代谢的影响及其作用机制.方法:雄性SD大鼠随机分成空白组6只和造模组18只,造模组予亚氨基二丙腈(IDPN)250 mg·kg-1·d-1,空白组予0.9%氯化钠溶液250 mg·kg-1·d-1,均腹腔注射7 d;造模组在腹腔注射基础上慢性束缚21 d后根据刻板评分结果进行二次分组,分为模型组,硫必利组和健脾止动汤组各6只,予相应药物灌胃28 d.比色法测定大鼠血清中葡萄糖(GLU)、丙酮酸(Pyruvate)、乳酸(LD)含量,HE染色观察大鼠海马组织形态,免疫组化法检测海马中葡萄糖转运蛋白1(GLUT1)、葡萄糖转运蛋白3(GLUT3)表达.结果:与空白组比较,模型组血清GLU、Pyruvate、LD水平均显著上升(P<0.05),海马组织细胞排列散乱疏松,可见细胞核萎缩、坏死,GLUT1、GLUT3蛋白表达显著升高(P<0.05).与模型组比较,健脾止动汤干预后血清Pyruvate、LD水平显著下降(P<0.05),海马组织病理损伤减轻,GLUT1、GLUT3蛋白表达显著降低(P<0.05).结论:健脾止动汤可保护脑组织,影响糖代谢及海马葡萄糖转运蛋白表达,其改善抽动的机制或与糖代谢重编程有关.
Study on the effect and mechanism of Jianpi Zhidong Decoction on glucose metabolism chronic in restraint stress Tourette syndrome model
Objective:To investigate the effects and its mechanism of Jianpi Zhidong Decoction on glucose metabolism in restraint stress Tourette syndrome(TS)rat model.Methods:Male SD rats were randomly divided into a blank group(n=6)and a model group(n=18).The model group was given IDPN(250 mg·kg-1·d-1)intraperitoneal injection for 7 days and chronic restraint for 21 days.The blank group was given normal saline(250 mg·kg-1·d-1)for 7 days.The model group was divided into three groups according to stereotype score,which were model group,Tiapride group and Jianpi Zhidong Decoction group,and gavaged with corresponding drug for 28 days.The content of glucose(GLU),Pyruvate(Pyruvate)and lactic acid(LD)in serum was detected by colorimetric method.The histopathological morphology of hippocampus was observed by hematoxylin eosin(HE).The expression of glucose transporter 1(GLUT1)and glucose transporter 3(GLUT3)in hippocampus was detected by immunohistochemical method.Results:Compared with blank group,the levels of GLU,Pyruvate and LD in model group were significantly increased(P<0.05),the cell arrangement of hippocampus was scattered and loose,the nucleus atrophy and necrosis were observed,and the expression of GLUT1 and GLUT3 protein was significantly increased(P<0.05).Compared with model group,the levels of GLU,Pyruvate and LD decreased after Jianpi Zhidong Decoction intervention,the pathological injury of hippocampus was alleviated,and the expression of GLUT1 and GLUT3 protein was significantly decreased(P<0.05).Conclusion:Jianpi Zhidong Decoction can protect brain tissue,affect glucose metabolism and the expression of glucose transporter in hippocampus,and may be related to the mechanism of glucose metabolism reprogramming.
