补肾温阳化瘀方调节SIRT1-FoxO1介导的细胞自噬治疗子宫内膜异位症的机制研究

目的:探讨SIRT1-FoxO1介导的细胞自噬在子宫内膜异位症(EMs)发展中的机制以及补肾温阳化瘀方治疗EMs的疗效与可能的作用机制.方法:将12Z细胞分为CON组、DMSO组(+同体积DMSO)及EX527组(+SIRT1抑制剂),采用划痕实验检测各组细胞的迁移能力,Transwell小室实验检测细胞侵袭能力,采用免疫荧光及Western Blot分别检测SIRT1、FoxO1及自噬相关因子蛋白定位及含量表达.将100只大鼠随机分为假手术组、模型组、中药组、中药+EX527组及EX527组.EMs造模成功后,中药组及中药+EX527组给予补肾温阳化瘀方灌胃,假手术组、模型组及EX527组同体积纯净水灌胃;中药+EX527组、EX527组给予腹腔注射EX527溶剂,其余各组给予腹腔注射同体积0.9%氯化钠溶液,连续3周.免疫组化与Western Blot分别检测SIRT1、Fox01及自噬相关因子蛋白定位及含量表达.结果:细胞实验中,与CON组或DMSO组比较,EX527组的迁移能力和侵袭能力明显增强,SIRT1水平显著下降(P<0.05),FoxO1及自噬相关因子水平显著升高(P<0.05,P<0.01);动物实验中,与假手术组比较,模型组与EX527组的SIRT1水平显著下降(P<0.05),FoxO1及自噬相关因子水平显著上升(P<0.01),中药组及中药+EX527组的SIRT1水平显著升高(P<0.01),FoxO1及自噬相关因子水平则显著降低(P<0.05,P<0.01).结论:SIRT1通过介导FoxO1负向调控细胞自噬水平,补肾温阳化瘀方可能通过上调SIRT1,抑制FoxO1介导的自噬,从而起到治疗EMs的作用.
Mechanism study on the regulation of SIRT1-FoxO1 mediated cellular autophagy in the treatment of endometriosis by Bushen Wenyang Huayu Recipe
Objective:To explore the mechanism of SIRT1 FoxO1 mediated autophagy in the development of endometriosis(EMs),as well as the therapeutic effect and possible mechanism of Bushen Wenyang Huayu Recipe in the treatment of EMs.Methods:12Z cells were divided into CON group,DMSO group(+DMSO of the same volume),and EX527 group(+SIRT1 inhibitor).The migration ability of each group of cells was detected using scratch assay,and the invasion ability of cells was detected using Transwell cell assay.After collection,the localization and content expression of SIRT1,FoxOl,and autophagy related factor proteins were detected using immunofluorescence and Western Blot,respectively.100 experimental rats were randomly divided into sham surgery group,model group,traditional Chinese medicine(TCM)group,TCM+EX527 group,and EX527 group.After successful modeling of EMs,the TCM group and the TCM+EX527 group were given the Bushen Wenyang Huayu Recipe by gavage,while the sham operation group,model group,and EX527 group were gavaged with the same volume of pure water;The TCM+EX527 group and EX527 group were given intraperitoneal injection of EX527 solvent,while the other groups were given intraperitoneal injection of the same volume of physiological saline for 3 consecutive weeks.Immunohistochemistry and Western Blot were used to detect the localization and content expression of SIRT1,FoxO1,and autophagy related factors,respectively.Results:In cell experiments,compared with the CON or DMSO groups,the EX527 group showed a significant increase in migration and invasion ability,a significant decrease in SIRT1 levels(P<0.05),and a significant increase in FoxO1 and autophagy related factors levels(P<0.05,P<0.01).In animal experiments,compared with the sham surgery group,the SIRT1 levels in the model group and EX527 group were significantly reduced(P<0.05),while the levels of FoxO1 and autophagy related factors were significantly increased(P<0.01).The SIRT1 levels in the TCM group and the TCM+EX527 group were significantly increased(P<0.01),while the levels of FoxO1 and autophagy related factors were significantly decreased(P<0.05,P<0.01).Conclusions:SIRT1 mediates FoxOl to negatively regulate cell autophagy levels.The Bushen Wenyang Huayu Recipe may upregulate SIRT1,inhibit FoxO1 mediated autophagy,and thus have a therapeutic effect on EMs.
