麻杏仙葶汤对湿热证肺炎幼龄大鼠的疗效机制研究

目的:探究麻杏仙葶汤治疗湿热证肺炎幼龄大鼠的疗效机制.方法:4~6周龄SD大鼠35只,随机分为空白组、肺炎组、湿热证肺炎组、地塞米松组和麻杏仙葶汤高、中、低剂量组.采用"高脂高糖饮食+湿热环境+腹腔注射脂多糖"方式制备湿热证肺炎幼鼠模型,并予相应药物灌胃治疗.观察幼鼠一般情况变化,检测其肺脏指数、肺组织病理学和血浆促炎因子IL-1β、IL-6、TNF-α含量.结果:湿热证肺炎幼龄大鼠模型制备成功.与空白组比较,湿热证肺炎组大鼠体温、肺脏指数显著升高(P<0.01),肺组织损伤严重,血浆IL-1β、IL-6、TNF-α含量显著升高(P<0.01).与湿热证肺炎组比较,麻杏仙葶汤中、高剂量组体温、肺脏指数显著降低(P<0.05,P<0.01);各剂量组大鼠肺组织损伤有不同程度减轻;中、高剂量组血浆IL-1β、IL-6、TNF-α含量显著降低(P<0.05,P<0.01).结论:麻杏仙葶汤能有效改善湿热证肺炎幼鼠的症状,抑制炎症反应,减轻肺组织损伤.
Study on therapeutic mechanism of Maxing Xianting Decoction on young rats with damp-heat syndrome of pneumonia
Objective:To investigate the mechanism of Maxing Xianting Decoction in treating young rats with damp-heat syndrome of pneumonia.Methods:Thirty-five SD rats of 4-6 weeks old were randomly divided into blank group,pneumonia group,damp-heat syndrome of pneumonia group,dexamethasone group and high-,medium-and low-dose groups of Maxing Xianting Decoction.The'high-fat and high-sugar diet+damp-heat environment+intraperitoneal injection of lipopolysaccharide'method was used to establish the damp-heat syndrome of pneumonia model of the young rats,and medications were given by gavage.Changes in the physical signs of young rats were observed,and lung organ index,lung histopathology and plasma levels of pro-inflammatory factors IL-1β,IL-6 and TNF-α were measured.Results:The young rat model of pneumonia with damp-heat syndrome was successfully prepared.Compared with the blank group,the body temperature and lung organ index of rats in damp-heat syndrome pneumonia group was significantly increased(P<0.01),the lung tissue was severely damaged,and the plasma levels of IL-1 β,IL-6 and TNF-α were significantly increased(P<0.01).Compared with damp-heat syndrome pneumonia group,the body temperature and lung organ index of rats in the medium-dose and high-dose groups of Maxing Xianting Decoction decreased most significantly(P<0.05,P<0.01),the lung tissue damage of rats in each dose group was alleviated to different degrees,and the plasma content of IL-1β,IL-6,TNF-α in the medium-dose and high-dose groups were significantly reduced(P<0.05,P<0.01).Conclusion:Maxing Xianting Decoction can effectively improve the symptoms of young rats with damp-heat syndrome of pneumonia,inhibit the inflammatory reaction,and reduce the lung tissue damage.
