清肺解毒颗粒调控Ⅰ型干扰素通路抗RSV效应的机制研究

目的:基于I型干扰素通路探究清肺解毒颗粒(QJG)发挥抗呼吸道合胞病毒(RSV)的效应机制.方法:将40只BALB/c雌性小鼠随机分为对照组(Con组),模型组(RSV组),RSV+清肺解毒颗粒组(RSV+QJG组),RSV+IFN-α 阻滞剂组(RSV+IFN-α Inh组),RSV+IFN-α 阻滞剂+清肺解毒颗粒组(RSV+IFN-α Inh+QJG组).除Con组外,其余各组小鼠给予RSV毒株,其中RSV+IFN-α Inh组、RSV+IFN-α Inh+QJG组在两次滴鼻造模前分别给予IFN-α阻滞剂.在感染后治疗组采用QJG给药治疗.观察各组小鼠的一般状态,并在治疗3d后处死取材.HE染色法观察肺组织的病理改变并评分,ELISA方法检测小鼠血清中IFN-α/β的水平,Q-PCR检测小鼠肺组织RIG-Ⅰ、MAVS、RSV-F、RSV-G等基因的转录水平,免疫荧光、Western Blot检测RSV-F、IRF3、OAS1、MX1等蛋白表达水平.结果:与RSV组比较,QJG药物治疗能够显著抑制肺组织RSV水平,同时改善肺组织炎性损伤(P<0.01).与RSV组比较,RSV+QJG组小鼠肺组织中RIG-1、MAVS、p-IRF3、IFN-α/β、OAS1、MX1 等基因的表达水平显著升高(P<0.01,P<0.05).IFN-α阻滞剂的干预能够影响QJG的抗病毒效应及IFN-α、OAS1、MX1升高水平(P<0.01,P<0.05).结论:QJG可能通过调控Ⅰ型干扰素通路从而发挥抗RSV的效应.
Study on anti-RSV mechanism of Qingfei Jiedu Granules in regulating type Ⅰinterferon pathway
Objective:To explore the effect mechanism of Qingfei Jiedu Granules(QJG)on respiratory syncytial virus(RSV)based on type Ⅰ interferon pathway.Methods:Forty female BALB/c mice were randomly divided into control group(Con),model group(RSV),RSV+Qingfei Jiedu Granules group(RSV+QJG),RSV+IFN-α blocker group(RSV+IFN-α Inh).RSV+IFN-α blocker+Qingfei Jiedu Granules(RSV+IFN-α Inh+QJG).Except for Con group,other groups were given RSV strain,and RSV+IFN-α Inh group and RSV+IFN-α Inh+QJG group were given IFN-α blocker before two nasal drops.After infection,the treatment group was treated with QJG.The general state of mice in each group was observed,and the mice were sacrificed for sampling after 3 days of treatment.The pathological changes of lung tissues were observed by HE staining and scored.The serum levels of IFN-α/β were detected by ELISA.The transcription levels of RIG-I,MAVS,IFN-α/β,RSVF,RSVG and other genes in lung tissues of mice were detected by Q-PCR.The protein expression levels of RSV-F,IRF3,OAS1 and MX1 genes were detected by immunofluorescence and Western Blot.Results:Compared with RSV group,QJG drug therapy could significantly inhibit the level of RSV in lung tissue and improve the inflammatory injury of lung tissue(P<0.0l).Compared with RSV group,the expression levels of RIG-I,MAVS,p-IRF3,IFN-α/β,OAS1 and MX1 genes in lung tissue of mice in RSV+QJG group were significantly increased(P<0.01,P<0.05).Intervention with IFN-α blockers could affect the antiviral effect of QJG and the elevated levels of IFN-α,OAS1,MX1 genes(P<0.01,P<0.05).Conclusion:QJG may exert anti-RSV effect by regulating type Ⅰ interferon pathway.
