Study on optimization of Box-Behnken response surface method extraction and chemical composition analysis of essential oil from Chimonanthus nitens Oliv.
Objective:To optimize the extraction conditions of essential oil from Chimonanthus nitens Oliv.by using Box-Behnken response surface methodology,and to study its chemical components by GC-MS.Methods:By designing Box-Behnken response surface model,the effects of crushing particle size,soaking time,extraction time and liquid material ratio on the extraction rate of Chimonanthus nitens Oliv.essential oil were investigated.Then the response surface model is used to fit the experimental results to determine the final optimal combination conditions,The chemical constituents of essential oil from Chimonanthus nitens Oliv.were analyzed by GC-MS.Results:The optimal technological conditions for extraction of Chimonanthus nitens Oliv.was as follows:The yield of essential oil can reach 7.60%under the conditions of crushing particle size of 24 mesh,soaking time of 1.4 h,extraction time of 3.1 h and liquid-material ratio of 31∶1 mL/g;A total of 56 compounds were extracted by optimized simultaneous distillation-extraction,including 12 alcohols,accounting for 47.98%,33 olefins,with The relative content of 25.11%,and other compounds,including esters,alkys and aldehydes,accounted for 7.82%.Conclusion:the optimized extraction process of Chimonanthus nitens Oliv.essential oil is stable and reliable.This study can provide a theoretical basis for the further development of Chimonanthus praecox essential oil products.
