基于DNA分子鉴定与化学模式识别方法的藏药索罗嘎布品种鉴别

摘要
目的:基于DNA分子鉴定和高效液相色谱(HPLC)指纹图谱结合化学模式识别方法对藏药索罗嘎布进行鉴别.方法:提取20批索罗嘎布药材的DNA,通过PCR扩增与测序筛选合适的引物序列,分析药材序列特征,并构建系统发育树;建立HPLC指纹图谱,进行相似度评价,并运用系统聚类分析(CA)、主成分分析(PCA)和正交偏最小二乘法-判别分析(OPLS-DA)进行化学模式识别研究.结果:基于ITS-18F/ITS-25R序列和HPLC指纹图谱均可有效地将20批索罗嘎布分为单花荠和丛菔属植物两类.结论:本研究所建立的方法可为藏药索罗嘎布的品种鉴别、质量提升与应用提供参考.
Abstract
Objective:To identify of Tibetan medicine Suoluogabu based on DNA molecular identification and HPLC fingerprinting combined with chemical pattern recognition methods.Methods:The DNA of 20 batches of Suoluogabu medicinal herbs was extracted,different primer sequences by PCR and sequenced was amplified,and the appropriate primer sequences was screened,the sequence characteristics of medicinal materials was analyzed and a phylogenetic tree was constructed.HPLC was used to establish the fingerprints of Suoluogabu,and similarity evaluation,cluster analysis(CA),principal pattern analysis(PCA),and orthogonal partial least squares-discriminant analysis(OPLS-DA)were applied to investigate the chemical pattern recognition.Results:The ITS-18F/ITS-25R sequence and HPLC fingerprint could effectively divide 20 batches Suoluogabu into two categories:gonococcus and conglomerate.Conclusion:The method established by this institute can provide a reference for the identification,quality improvement,and application of Tibetan medicine Suoluogabu.
