目的:探索正常小鼠针刺后血清外泌体对缺氧小鼠生存时间的影响.方法:将KM种小鼠随机分为空白对照组、电针组,电针预处理后即刻复制缺氧模型,以生存时间为观察指标;将KM种小鼠随机分为E0、E1、E4、E7组,运用ExoQuick法提取针刺后外泌体(Acu-exo)进行鉴定.将Acu-exo分别回输至回输E0组、回输E1组、回输E4组、回输E7组,30min后复制缺氧模型,以生存时间为观察指标;将KM种小鼠随机分为空白对照组、电针组、电针+DMSO组、电针+GW4869组,其中电针+DMSO组、电针+GW4869组分别进行腹腔注射DMSO及鞘磷脂酶抑制剂GW4869,1 h后,将小鼠进行电针预处理后即刻进行缺氧模型复制,以生存时间为观察指标.结果:电针组较空白对照组的生存时间显著提高(P<0.01);回输E1组、回输E4组、回输E7组的耐缺氧时间较空白对照组显著升高(P<0.01);电针组较空白对照组和电针+GW4869组生存时间显著提高(P<0.01).结论:回输正常小鼠针刺后血清Acu-exo可提高缺氧小鼠的生存时间,为针刺治未病及基础研究转化应用提供一定依据.
Preliminary study on the effects of serum exosome on the survival time of hypoxic mice after electroacupuncture in normal mice and its action law
Objective:To investigate the effects of serum exosome on the survival time of hypoxic mice after acupuncture in normal mice.Methods:KM mice were randomly divided into blank control group and electroacupuncture group.The hypoxia model was replicated immediately after electroacupuncture pretreatment,and the survival time was observed.KM mice were randomly divided into E0,E1,E4 and E7 groups,and acupuncture-exosome(Acu-exo)was extracted by ExoQuick method for identification.Acu-exo was infused back to reinfusion E0 group,reinfusion E1 group,reinfusion E4 group and reinfusion E7 group,respectively.After 30 min,the hypoxia model was replicated,and the survival time was observed.KM mice were randomly divided into blank control group,electroacupuncture group,electroacupuncture+DMSO group and electroacupuncture+GW4869 group.The mice in electroacupuncture+DMSO group and electroacupuncture+GW4869 group were given intraperitoneal injection of DMSO and GW4869,respectively.After 1 hour,the mice were pretreated with electroacupuncture and the hypoxia model was replicated immediately.The survival time was observed.Results:The survival time of electroacupuncture group was significantly longer than that of blank control group(P<0.01).Compared with the blank control group,the hypoxia tolerance time of reinfusion E1 group,reinfusion E4 group and reinfusion E7 group was significantly improved(P<0.01).Compared with the blank control group and electroacupuncture+GW4869 group,the survival time of the electroacupuncture group was significantly improved(P<0.01).Conclusion:Reinfusion of Acu-exo can improve the survival time of hypoxic mice,and provide a basis for the basic research and application of acupuncture and preventive treatment of disease.
