Protective effect of Epimedii Folium and Curculiginis Rhizoma on osteoblasts injury induced by high glucose via regulating lncRNA Gabpb1-AS1
Objective:To investigate the protective mechanism of Epimedii Folium and Curculiginis Rhizoma(EBCOs)against osteoblast injury induced by high glucose concentration at the lncRNA level.Methods:EBCOs was extracted from dried leaves of Epimedii Folium and dried rhizomes of Curculiginis Rhizoma.Osteoblasts were divided into three groups:control group(1.65 mmol/L glucose treatment for 24 h),HG model group(33.1 mmol/L glucose treatment for 24 h)and EBCO group(33.1 mmol/L glucose treatment for 24 h and 10 μg/mL EBCOs-containing serum intervention).The lncRNA expression level was detected by lncRNA sequencing.Expressions of lncRNA Gabpb1-AS1 and Nrf2 mRNA were detected by qRT-PCR.Cell transfection silenced lncRNA Gabpb1-AS1.The protein expression of Keap1,Nrf2 and BMP2 were detected by Western Blot.Results:The lncRNA Gabpb1-AS1 in HG group was significantly increased compared with control group(P<0.01),and decreased after EBCOs intervention(P<0.01).After silencing the lncRNA Gabpb1-AS1 by transfection,the lncRNA Gabpb1-AS1 was significantly decreased(P<0.01),the protein expressions of Nrf2 and BMP2 were significantly increased while the protein expressions of Keap1 were significantly decreased(P<0.01).Conclusion:EBCOs may relief high glucose-induced damage in osteoblasts by down-regulating lncRNA Gabpb1-AS1.
