健骨颗粒正丁醇极性部位对去卵巢大鼠OPG/RANKL的影响

目的:观察健骨颗粒正丁醇极性部位对去卵巢大鼠骨保护素(OPG)和核因子-κB配体受体致活剂(RANKL)的影响,探讨其治疗绝经后骨疏松症(PMOP)的作用机制.方法:切除雌性SD大鼠双侧卵巢制造PMOP模型,运用双能X线骨密度仪测定骨密度(BMD);ELISA法检测血清中雌二醇(E2)、抗酒石酸酸性磷酸酶(TRAP)、OPG和RANKL的含量,并计算OPG/RANKL比值;Western Blot法测定骨组织中TRAP、OPG和RANKL蛋白表达,并计算OPG/RANKL比值.结果:与生理盐水组比较,正丁醇组术后第12周BMD显著增高(P<0.01),术后第8、12周血清E2和OPG水平显著升高(P<0.01);术后8周TRAP在血清中的含量和在骨组织中的蛋白表达均显著降低(P<0.01);术后第8周骨组织OPG蛋白表达显著升高(P<0.01);术后第4、8周骨组织中RANKL蛋白表达均显著降低(P<0.01);术后3个时间点血清中OPG/RANKL比值都显著升高(P<0.01).结论:健骨颗粒正丁醇极性部位可调节OPG与RANKL之间的代谢平衡,提高血清中OPG/RANKL的比值,降低骨吸收,改善去卵巢大鼠骨骼的代谢水平,提高BMD.
Effects of Jiangu Granules n-butanol polar fraction on OPG/RANKL in ovariectomized rats
Objective:To observe the effect of Jiangu Granules n-butanol polar fraction on osteoprotegerin(OPG)and receptor activator of nuclear factor-KB ligand(RANKL)in ovariectomized rats,and to explore its mechanism in the treatment of postmenopausal osteoporosis(PMOP).Methods:The model was established by ovariectomy in female SD rats.Bone mineral density(BMD)was measured by dual energy X-ray absorptiometry.E2,TRAP,OPG,RANKL were detected by ELISA,and calculated the OPG/RANKL ratio.The contents of TRAP,OPG and RANKL proteins in bone tissue were measured by Western Blot,and calculated the OPG/RANKL ratio.Results:Compared with normal saline group,BMD in n-butanol group at 12 weeks after surgery was significantly increased(P<0.01),serum E2 and OPG levels at 8 and 12 weeks after surgery were significantly increased(P<0.01);the content of TRAP in serum and protein expression in bone tissue were significantly decreased at 8 weeks after operation(P<0.01);the expression of OPG protein in bone tissue was significantly increased at 8 weeks after operation(P<0.01);the expression of RANKL protein in bone tissue was significantly decreased at 4 and 8 weeks after operation(P<0.01);the OPG/RANKL ratio in serum was significantly increased at three time points after operation(P<0.01).Conclusion:The n-butanol polar fraction of Jiangu Granules can regulate the metabolic balance between OPG and RANKL,increase the ratio of OPG/RANKL in serum,reduce bone resorption,improve the metabolic level of rat bone after oophorectomy and increase BMD.
