Effects of Bushen Tiaogan Jiangzhuo Formula on white fat browning in obese polycystic ovary syndrome rats
Objective:To explore the possible mechanism of Bushen Tiaogan Jiangzhuo Formula in the treatment of obese polycystic ovary syndrome(PCOS).Method:Sixty 6-week-old SPF grade SD female rats were divided into blank group(10 rats)and marking model group(50 rats).The making model group was fed with letrozole by gavage and a high-fat diet to prepare an obese PCOS rat model.The 50 rats that successfully established the model were randomly divided into model group,low,medium and high dose group of Bushen Tiaogan Jiangzhuo Formula,and metformin group.After the administration,the ELISA method was used to detect serum leptin(LP)and adiponectin(ADPN);Western Blot and RT-qPCR were used to detect the protein and mRNA expression of AMPK,PGC-1 α,and UCP-1 in white adipose tissue(WAT)and brown adipose tissue(BAT)of rats.Results:Compared with the blank group,LP increased in the model group and ADPN decreased(P<0.01);the LP of the low,medium,and high dose group of Bushen Tiaogan Jiangzhuo Formula was significantly lower than that of the model group(P<0.01),while the ADPN of the medium and high dose groups of Bushen Tiaogan Jiangzhuo Formula was significantly increased(P<0.05).Compared with the model group,the medium dose group of Bushen Tiaogan Jiangzhuo Formula could activate the WAT AMPK/PGC-1 α pathway and increase the expression of UCP-1(P<0.01);Metformin also activated the AMPK/PGC-1 α pathway(P<0.05,P<0.01).Compared with the control group,detection of BAT revealed a decrease in the mRNA and protein expression of AMPK,PGC-1 α,and UCP-1 in the model group(P<0.05,P<0.01);the mRNA and protein expression of AMPK,PGC-1 α and UCP-1 in the medium dose group of Bushen Tiaogan Jinagzhuo Formula significantly increased(P<0.01,P<0.05).Conclusion:Bushen Tiaogan Jiangzhuo Formula can activate the AMPK/PGC-1 α pathway,increase the expression of UCP-1,reduce LP,increase ADPN levels,promote WAT browning,activate BAT,restore normal lipid metabolism,reduce FT,and restore ovulation.
