Study on four natures characterization of Lubian based on AMPK/SIRT1/PGC-1 α pathway
Objective:To study the pharmacological characterization of Lubian and explore its four qi attribution.Methods:Eighty-eight healthy male Kunming mice were randomly divided into blank group,deficiency cold model group,ginseng group,deficiency cold plus low,medium and high dose Lubian groups,deficiency heat model group,American ginseng group,and deficiency heat plus low,medium and high dose Lubian groups after 1 week of adaptive feeding.Each group had 8 mice,totaling 11 groups.Each afternoon,hydrocortisone and dexamethasone acetate were administered by gavage to prepare models of deficiency cold and deficiency heat.The corresponding drugs were given at regular intervals every morning,while the blank group was given blank solvent for 14 consecutive days.Measure the exhaustion swimming time of mice in each group 30 minutes after administration on the 14th day;On the 15th day,blood was extracted from eyeballs and serum was separated to determine the levels of blood lactate(LA),urea nitrogen(BUN),total cholesterol(TC),triglycerides(TG)in the serum.Thigh skeletal muscles were dissected to determine muscle glycogen levels Na+-K+-ATPase,Ca2+-Mg2+-ATPase activity,as well as the protein expression status of P-AMPK,AMPK,SIRT1,PGC-1 α in muscle tissue.Results:Compared with the deficiency cold model group,the deficiency cold plus low,medium dose Lubian groups showed a significant increase in body mass,improved deficiency cold symptoms,significantly increased exhaustion swimming time,significantly decreased blood LA content,significantly increased BUN,TC and muscle glycogen content,and significantly increased muscle Na+-K+-ATPase and Ca2+-Mg2+-ATPase activities,muscle tissue P-AMPK/AMPK values,SIRT1,PGC-1 α content significantly increased.Compared with the deficiency heat model group,the deficiency heat plus low,medium dose Lubian groups showed a significant increase in body mass,improvement in deficiency heat symptoms,a significant increase in exhausted swimming time,a significant decrease in blood LA and BUN,a significant increase in muscle glycogen content,the activity of muscle and Na+-K+-ATPase and Ca2+-Mg2+-ATPase was significantly reduced,and the P-AMPK/AMPK values,SIRT1,and PGC-1 αin muscle tissue were significantly reduced.Conclusion:Lubian can affect AMPK/SIRT1/PGC-1 α pathway to regulate the abnormal state of energy metabolism in muscle tissue,thereby bi-directional regulating the state of deficiency cold and deficiency heat in mice,enhancing the physical strength of mice with deficiency cold and deficiency heat.Therefore,the four qi of Lubian belongs to ping.
