Study on mechanism of Qianliexiao Decotion regulating the inflammatory response of experimental autoimmune prostatitis through IL-6/JAK2/STAT3 pathway
Objective:To study the mechanism of Qianliexiao Decoction regulating the inflammatory response of experimental autoimmune prostatitis(EAP)through IL-6/JAK2/STAT3 pathway.Methods:A total of 180 male SPF-grade C57BL/6 mice were randomly divided into blank group,model group,positive control group(AG490 inhibitor),low/medium/high dose Qianliexiao Decoction group.EAP mouse models with damp heat syndrome were prepared by using complex factors,and evaluated by prostate wet weight index and HE staining pathological model.Western Blot and RT-PCR were used to detect IL-6,gp130,JAK2,STAT3,SOCS3,RORγ t,IL-17A,Foxp3 protein and gene expression level in prostate tissues of EAP mice.Results:The prostate wet weight index in the model group was higher than that in the blank group(P<0.05).Compared with the blank group,the model group showed infiltration of monocytes and inflammatory cells,thickening of acinar cell wall,and disorder of glandular cavity structure.Each drug group could improve the degree of inflammatory infiltration of the prostate of model mice to varying degrees.The intervention effect of Qianliexiao Decoction in each dose group was most significant in the high-dose group.Compared with the EAP group,except ROR γ t protein in low dose Qianliexiao Decoction group,the Qianliexiao Decoction groups down-regulated IL-6,gpl30,JAK2,STAT3,SOCS3,RORγt,IL-17A,Foxp3 protein expression(P<0.01,P<0.05).Conclusion:Qianliexiao Decoction can inhibit the experimental autoimmune prostatitis reaction through IL-6/JAK2/STAT3 pathway,thereby improving the pathological structure and autoimmune reaction function of prostate in EAP mice.
