Objective:To verify the intervention effects of Tuina on opioid-induced constipation(OIC),to observe the effects of Tuina on the TRPV1,β-arrestin2,and μ-opioid receptors(MDR)in intestinal tissue of OIC rat model.Methods:Fifty SD rats were randomly divided into blank group,OIC group,OIC+Tuina group,TRPV1 blocker group,OIC+TRPV1 blocker+Tuina group,10 rats in each group.The OIC rat model was prepared by continuous injection of Morphine,and the intervention was performed by rubbing the abdomen clockwise to observe its intervention effect.The protein and mRNA expressions of TRPV1,β-arrestin2 and MOR in intestinal tissues were detected by Western Blot and RT-PCR.Results:Compared with the OIC group,the number of feces particles,wet weight of feces,and water content of feces in the OIC+Tuina group were significantly improved(P<0.05,P<0.01);Tuina could up-regulate the expression of TRPV1 in the intestinal tissue of OIC rat model(P<0.05),the number of feces particles and water content in feces of rats in the OIC+Tuina+TRPV1 blocker group increased(P<0.05),but the wet weight of feces was not statistically significant.Conclusion:Tuina can relieve OIC;the regulatory mechanism of Tuina on TRPV1,β-arrestin2,and MOR in the intestinal tissue of OIC rat model still needs further exploration.
