基于16S rDNA测序研究茱萸丸对动脉粥样硬化小鼠肠道菌群结构的影响

目的:观察茱萸丸对ApoE-/-动脉粥样硬化(AS)小鼠肠道菌群的影响.方法:采用高脂饲料喂养ApoE-/-小鼠诱导AS模型,总造模周期为12周.造模成功的50只ApoE-/-小鼠随机分为模型组,茱萸丸低、中、高剂量组,阿托伐他汀钙组,每组10只,C57BL/6J小鼠10只作为空白对照组.造模后空白对照组与模型组给予等体积无菌蒸馏水灌胃,茱萸丸低、中、高剂量组分别给予茱萸丸混悬液130.54、261.08、552.16 mg/kg灌胃,阿托伐他汀钙组给予阿托伐他汀钙混悬液10.40 mg/kg灌胃,各组小鼠均灌胃12周.检测小鼠体质量、摄食量;苏木素-伊红(HE)染色观察小鼠主动脉组织的病理学改变;酶联免疫吸附测定法(ELISA)检测血清中氧化型低密度脂蛋白(ox-LDL)、单核细胞趋化蛋白-1(MCP-1)、血管细胞黏附分子-1(VCAM-1)、细胞间黏附分子-1(ICAM-1)水平;采用16S rDNA技术检测小鼠粪便的肠道菌群Alpha、Beta多样性及物种组成差异.结果:与空白对照组比较,模型组小鼠体质量、摄食量降低;HE染色可见主动脉内膜大面积增厚;血清ox-LDL、MCP-1、VCAM-1、ICAM-1水平显著升高(P<0.01);Chao1指数、Ace指数以及observed species指数显著降低(P<0.01);疣微菌门、杜氏牙形石属、乳杆菌属相对丰度显著升高(P<0.01),拟杆菌门、放线菌门、圣埃里立支杆菌属、粪杆菌属、布劳特氏属相对丰度显著降低(P<0.01).与模型组比较,各给药组体质量、摄食量上升;HE染色可见随着茱萸丸浓度的增加,斑块富集区域逐渐缩小;血清ox-LDL、MCP-1、VCAM-1、ICAM-1水平显著下降(P<0.01);Chao1指数、Ace指数以及observed species指数显著升高(P<0.01);拟杆菌门、变形菌门、粪杆菌属、布劳特氏属相对丰度显著升高(P<0.01),厚壁菌门、放线菌门、疣微菌门、杜氏牙形石属、圣埃里立支杆菌属、乳杆菌属相对丰度显著降低(P<0.01).结论:茱萸丸能够显著抑制AS小鼠主动脉病理改变,调节肠道菌群中有益菌和有害菌的相对丰度,纠正菌群紊乱情况,从而减轻主动脉炎症反应.
Effects of Zhuyu Pills on intestinal flora structure of atherosclerosis mice based on 16S rDNA sequencing
Objective:To observe the effect of Zhuyu Pills on intestinal flora of ApoE-/-atherosclerosis(AS)mice.Methods:The mouse model of AS was established by feeding with a high-fat diet for 12 weeks.The 50 successful modeled ApoE-/-mice were randomly divided into model group,low,medium and high dose groups of Zhuyu Pills and atorvastatin calcium group,with 10 mice in each group.Ten C57BL/6J mice were selected as normal group.After modeling,normal group and model group were given equal volume of sterile distilled water by gavage,low,medium and high dose groups were given 130.54,261.08 and 552.16 mg/kg by gavage,respectively,and atorvastatin calcium group was given 10.40 mg/kg atorvastatin calcium suspension by gavage,all of which were given gavage for a total of 12 weeks.The body weight and food intake of mice were detected.Hematoxylin-eosin(HE)staining was used to observe the pathological changes of aorta in mice.The levels of oxidized low-density lipoprotein(ox-LDL),monocyte chemotactic egg-1(MCP-1),vascular cell adhesion-1(VCAM-1)and intercellular adhesion molecule-1(ICAM-1)in serum were detected by enzyme-linked immunosorbent assay(ELISA).16S rDNA technique was used to detect the diversity of Alpha and Beta,and species composition of intestinal flora in mouse feces.Results:Compared with normal group,body weight and food intake of mice in model group decreased.HE staining showed large area thickening of the aortic intima.Serum ox-LDL,MCP-1,VCAM-1 and ICAM-1 levels were increased significantly(P<0.01).Chao1 index,Ace index and observed species index decreased(P<0.01).The relative abundance of Verrucomicrobiota,Dubosiella and Lactobacillus increased(P<0.01),while the relative abundance of Bacteroidota,Actinobacteria,Ligilactobacillus,Faecalibaculum and Blautia decreased significantly(P<0.01).Compared with the model group,the body weight and food intake of each administration group increased.HE staining showed that with the increase of the concentration of Zhuyu Pills,the plaque enrichment area decreased gradually.Serum ox-LDL,MCP-1,VCAM-1 and ICAM-1 were decreased significantly(P<0.01);Chao1 index,Ace index and observed species index were increased significantly(P<0.01).The relative abundance of Bacteroidota,Proteobacteria,Faecalibaculum and Blautia increased significantly(P<0.01),while Firmicutes,Actinobacteria,Verrucomicrobiota,Dubosiella,Ligilactobacillus and Lactobacillus decreased significantly(P<0.01).Conclusion:Zhuyu Pills can significantly inhibit the pathological changes of aorta in AS mice,regulate the relative abundance of beneficial and harmful bacteria in intestinal flora,correct the flora disorder,and thus reduce the inflammatory response of aorta.
