Pharmacological study on the effects of Jingfang Granules on promoting blood circulation and resolving stasis
Objective:To explore the effects of Jingfang Granules on promoting blood circulation and resolving stasis.Methods:Establish three zebrafish cardiovascular disease models,using ferric chloride,terfenadine,and arachidonic acid to establish zebrafish tail artery stenosis model,heart injury model,and thrombus model,respectively.Administer Jingfang Granules sample solution for intervention,and observe its effects on the diameter of the tail artery,the distance between the cardiac venous sinus and the arterial bulb(SV-BA),and the stained area of cardiac red blood cells;An acute blood stasis model was established in rats using adrenaline combined with ice water stress.The expression of four coagulation parameters(APTT,TT,PT,FIB),as well as vascular active substances(PGI2,t-PA,PAI,TXA2)and adhesion molecules(VCAM-1,ICAM-1)in the serum were detected.Results:The experimental results of zebrafish showed that Jingfang Granules relaxed the blood vessels in the tail of zebrafish(P<0.01)and significantly improved vascular stenosis;Increase the stained area of cardiac red blood cells(P<0.05,P<0.01)and inhibit thrombus formation;Although there is a certain decreasing trend in SV-BA,and there is no significant reversal effect on zebrafish heart injury.In addition,the results of pharmacological experiments in rats showed that Jingfang Granules can improve four coagulation indicators,as well as the expression of vascular active substances and adhesion molecules related to blood stasis.In terms of coagulation,Jingfang Granules can significantly reduce FIB content(P<0.01)and increase APTT,PT,and TT(P<0.05);In terms of factor detection,Jingfang Granules can increase the levels of PGI2 and t-PA(P<0.05),reduce the levels of TXA2,PAI,and VCAM-1(P<0.05).Conclusion:Jingfang Granules can promote blood circulation and resolving stasis,which may play the effect of promoting blood circulation and removing blood stasis by dilating blood vessels,anti-thrombosis,improving blood coagulation function and regulating the expression of vasoactive substances and adhesion molecules.
