基于特征图谱及高效液相色谱波长切换法同时测定当归饮子中6种成分

目的:建立当归饮子物质基准特征图谱和6种成分含量测定方法,以期为当归饮子物质基准的质量评价研究提供参考.方法:使用不同批次饮片制备15批当归饮子物质基准,利用高效液相色谱波长切换法建立当归饮子特征图谱,并进行特征峰归属和相似度分析;以绿原酸、芍药苷、阿魏酸、升麻素苷、5-O-甲基维斯阿米醇苷、甘草酸铵6种成分作为指标性成分建立当归饮子多成分含量测定方法,检测波长依次是绿原酸327 nm、升麻素苷254 nm、芍药苷230 nm、阿魏酸316 nm、5-O-甲基维斯阿米醇苷254 nm、甘草酸铵237 nm,流速为1.0 mL/min,柱温为30℃,进样量为10 μL.结果:15批当归饮子物质基准特征图谱共有9个特征峰,共指认出绿原酸、芍药苷、阿魏酸、升麻素苷、5-O-甲基维斯阿米醇苷、甘草酸铵6个色谱峰,相似度均>0.95;精密度、稳定性(24h)、重复性试验的RSD均<2%(n=6);平均加样回收率均>98%(RSD<2%,n=9).结论:建立的当归饮子物质基准特征图谱和含量测定方法简单易行,结果准确,重复性稳定性良好,可为当归饮子物质基准的质量评价提供参考.
Simultaneous determination of 6 components in Danggui Yinzi based on characteristic map and HPLC wavelength switching method
Objective:To establish the characteristic chromatogram of the reference substance of Danggui Yinzi and the content determination method of 6 components in Danggui Yinzi,in order to provide reference for the quality evaluation of the reference material of the Danggui Yinzi.Methods:Using different batches of decoction pieces to prepare 15 batches of Danggui Yinzi material benchmark,the characteristic chromatogram of Danggui Yinzi was established by HPLC wavelength switching method,and the characteristic peak attribution and similarity analysis were carried out.The six components of chlorogenic acid,paeoniflorin,ferulic acid,prim-O-glucosylcimifugin,5-O-methylvisammioside and ammonium glycyrrhetate were used as index components to establish a method for the determination of multi-component content in Danggui Yinzi.The detection wavelengths were 327 nm for chlorogenic acid,254 nm for prim-O-glucosylcimifugin,230 nm for paeoniflorin,316 nm for ferulic acid,254 nm for 5-O-methylvisammioside and 237 nm for ammonium glycyrrhizinate.The flow rate was 1.0 mL/min.The column temperature was set at 30 ℃,and sample size was 10 μL.Results:There were 9 characteristic peaks in the characteristic chromatograms of 15 batches of Danggui Yinzi,and 6 chromatographic peaks of chlorogenic acid,paeoniflorin,ferulic acid,prim-O-glucosylcimifugin,5-O-methylvisammioside and ammonium glycyrrhetate were identified.The similarity was more than 0.95.The RSD of precision,stability(24 h)and repeatability were less than 2%(n=6).The average spiked recoveries were all>98%(RSD<2%,n=9).Conclusion:The established characteristic spectrum and content determination method of Danggui Yinzi substance benchmark are simple,accurate and reproducible,which can provide reference for the quality evaluation of Danggui Yinzi substance benchmark.
